摘 要:挖掘抗病基因可为苹果抗病育种提供基因材料。本研究结合生物信息学分析、基因表达分析和功能验证,分析了长春花类受体激酶-1类蛋白(Catharanthus roseus receptor-like kinase-1-like,CrRLK1L)亚家族成员MdFER1在苹果腐烂病抗性中的作用。结果表明,MdFER1与拟南芥FER同源但序列相似性较低,仅为46.4%。在‘烟富6号’果实和‘王林’悬浮细胞中瞬时表达MdFER1,果实和细胞对苹果腐烂病的抗性显著降低。MdFER1瞬时表达可显著诱导活性氧(reactive oxygen species,ROS)相关基因TaNOX和超敏反应(hypersensitive response,HR)相关基因HSR203的表达,但抑制防御相关基因PR1和EDS1的表达。综上所述,MdFER1负调控‘烟富6号’果实和‘王林’悬浮细胞对苹果腐烂病抗性,ROS和HR信号被激活和防御相关基因表达被抑制是该基因负调控苹果腐烂病抗性的重要原因。Screening for resistance genes can provide valuable genetic materials for breeding apple varieties with enhanced resistance.We analyzed the role of MdFER1,a member of the Catharanthus roseus receptor-like kinase-1-like(CrRLK1L)proteins subfamily,in apple Valsa canker resistance through bioinformatics analysis,expression analysis,and functional verification of the gene.The results showed that MdFER1 is homologous to Arabidopsis FER,although the sequence similarity is only 46.4%.Transient expression of MdFER1 in either‘Yanfu 6’fruits or‘Wanglin’suspension cells significantly reduced resistance to apple Valsa canker.Additionally,MdFER1 transient expression significantly activated the expression of genes associated with reactive oxygen species(TaNOX)and hypersensitive response(HSR203),but inhibited the expression of defense-related genes PR1 and EDS1.In summary,MdFER1 negatively regulates resistance to apple Valsa canker in‘Yanfu 6’fruits or‘Wanglin’suspension cells.The activation of ROS and HR signaling,along with the inhibition of defense-related gene expression,are key factors contributing to the negative regulation of apple Valsa canker resistance by MdFER1.
