摘 要:目的建立以对照药材为对照物质的定性和不依赖多种对照品定量的吴茱萸药材“质-量”双标控制方法。方法采用高效液相色谱(HPLC)技术,以对照药材为对照物质,建立吴茱萸特征图谱,并通过质谱技术鉴定共有峰化学成分,通过评价对照药材与供试药材特征图谱相似度,判别药材真伪,即“质”;以吴茱萸碱作为基准物质进行准确定量,以吴茱萸碱为基准,计算其余特征峰相对含量,并采用特性峰的相对含量下限来判定吴茱萸药材优劣,即“量”。结果建立的吴茱萸特征图谱以及吴茱萸含量测定的方法符合方法学考察要求;鉴定3个共有色谱峰,分别为柠檬苦素、吴茱萸碱、吴茱萸次碱;不同产地供试药材与吴茱萸对照药材相比相似度均大于0.970;规定了吴茱萸特征峰化学成分相对含量下限。结论该方法不依赖多种对照品,可以快速简便的判别药材真伪,同时达到全面控制吴茱萸药材质量的目的。Objective To establish a"quality-quantity"double standard control method for Evodia officinalis,which is based on the quality of the control material and does not depend on the quantity of the control material.Methods The characteristic spectrum of Evodia officinalis was established by high performance liquid chromatography(HPLC),and the common peak chemical components were identified by mass spectrometry.The authenticity of Evodia officinalis was determined by evaluating the similarity of the characteristic spectrum of Evodia officinalis and the tested medicinal materials.The relative content of other characteristic peaks was calculated with evodiamine as the reference material,and the lower limit of relative content of characteristic peaks was used to judge the quality of evodiamine,that is,the quantity.Results The characteristic map of Euodiae Fructus and the determination method of Euodiae Fructus content were in line with the requirements of methodology investigation.Three chromatographic peaks were identified,namely limonin,evodiamine and rutaecarpine.The similarity between the tested herbs from different origin and the control herbs of Euodiae Fructus was greater than 0.970.The lower limit of relative content of chemical components in characteristic peak of Euodiae Fructus was defined.Conclusion This method can distinguish the authenticity of medicinal materials quickly and easily without relying on many reference materials,and achieve the purpose of controlling the quality of medicinal materials of Euodiae Fructus.
