摘　　要：目的:建立同时测定淡竹叶中荭草苷和异荭草苷含量的UPLC法,并采用该方法检测不同产地淡竹叶药材中荭草苷和异荭草苷含量。方法:淡竹叶经70%甲醇超声处理后采用Waters CORTECS T3色谱柱(2.1 mm×150 mm, 1.6μm),乙腈-0.2%磷酸溶液为流动相,梯度洗脱,流速为0.25 mL/min,柱温为40℃,检测波长为348 nm,分别对专属性、线性关系、重复性、中间精密度、准确性、耐用性等进行考察。结果:荭草苷和异荭草苷分别在选定的范围内线性关系良好(r≥0.999 9),平均加标回收率分别为100.10%、100.63%,RSD值分别为2.21%、2.40%(n=9),15批淡竹叶药材中荭草苷含量范围为0.002 3%~0.277 3%,异荭草苷含量范围为0.007 4%~0.339 6%。结论:所建方法准确稳定、重复性好,可用于淡竹叶原料质量控制,不同采收时间淡竹叶药材中荭草苷和异荭草苷含量差异较大,以四川、贵州产地较准,每年6-7月份采收的药材含量较高。Objective:To establish a UPLC method for the simultaneous determination of orientin and isoorientin in Lophatherum Gracile Brongn,and use this method to detect the contents of orientin and isoorientin in Lophatherum Gracile Brongn from different origins.Methods:Lophatherum Gracile Brongn were treated with 70%methanol ultrasound and then used a Waters CORTECS T3 column(2.1mm×150mm,1.6μm),with acetonitrile 0.2%phosphoric acid solution as the mobile phase,gradient elution,flow rate of 0.25mL/min,column temperature of 40℃,detection wavelength of 348nm.The specificity,linear relationship,reproducibility,intermediate precision,accuracy,and durability were investigated.Results:There was a good linear relationship between orientin and isoorientin within the selected range(r≥0.9999),with average spiked recoveries of 100.10%and 100.63%,and RSD values of 2.21%and 2.40%(n=9),respectively.The content of orientin in 15 batches of Lophatherum Gracile Brongn medicinal materials ranged from 0.0023%to 0.2773%,and the content of isoorientin ranged from 0.0074%to 0.3396%.Conclusion:This method is accurate,stable,and has good repeatability,and can be used for quality control of Lophatherum Gracile Brongn raw materials.The content of orientin and isoorientin in Lophatherum Gracile Brongn medicinal materials varies greatly at different harvesting times.The content of medicinal materials collected from June to July in Sichuan and Guizhou provinces is relatively high.
