摘 要:目的:基于阿尔茨海默病(AD)小鼠模型观察细叶远志皂苷(Tenuifolin)对小鼠一般状态及肝肾的影响。方法:40只雄性阿尔茨海默模型小鼠(APPswe/PSEN1dE9双转基因小鼠,APP/PS1小鼠)随机分为4组:模型组、细叶远志皂苷低、中、高剂量组,每组10只。另取10只同系野生型C57BL/6J小鼠作为正常对照组。各剂量组灌胃给予细叶远志皂苷低、中、高剂量(20 mg/kg、40 mg/kg、80 mg/kg),正常对照组及模型组小鼠灌胃给予同体积0.3%羧甲基纤维素钠溶液,1次/d,连续给药90 d。给药期间观察小鼠一般状态,每周测一次体质量。末次给药后处死小鼠,取肝脏及肾脏称重并计算肝脏系数及肾脏系数;HE染色观察小鼠肝肾组织病理变化。结果:与正常对照组相比,细叶远志皂苷各剂量组小鼠状态无明显异常,体质量、肝脏及肾脏系数未见显著变化(P>0.05);与正常对照组相比,细叶远志皂苷各剂量组肝肾组织无明显形态学变化。结论:细叶远志皂苷对APP/PS1小鼠基本无毒,不引起肝肾损伤,安全性较高。Objective:To investigate the toxicity of tenuifolin to liver and kidney in Mouse Models of Alzheimer's Disease.Methods:Forty male APP/PS1 double transgenic mice were randomly assigned into model group and tenuifolin low,medium,high dose groups with 10 mice in each group.10 wildtype C57BL/6J mice were used as normal controls.Mice were given tenuifolin(20mg/kg,40 mg/kg,and 80 mg/kg)or 0.3%sodium carboxymethylcellulose solution by gavage once a day for 90 consecutive days.The general physical signs were observed daily,and body weight was measured weekly.The liver and kidneys of mice were weighed,and the organ index was measured at the end of administration.Hematoxylin eosin(HE)staining was used to detect liver and renal pathological changes.Results:No obvious abnormal behavior of mice in each dose group was observed compared with the normal control group.Tenuifolin did not affect body weight,liver index,and kidney index(P>0.05).Histopathological examination showed no significant changes in the liver and kidney tissues of mice in the tenuifolin groups.Conclusion:No obvious toxic and side effects of tenuifolin were observed in liver and kidney of APP/PS1 double transgenic mice and it has a high degree of safty.
