摘 要:目的:探讨玉泉丸调控microRNA-320对糖尿病小鼠糖代谢的影响并阐明其作用机制。方法:生化分析仪检测各组小鼠血糖、甘油三酯、总胆固醇、血肌酐和尿素;RT-PCR检测microRNA-320、PI3K、Akt以及GLUT-1基因表达水平;Western Blot检测PI3K、Akt以及GLUT-1蛋白表达水平。结果:玉泉丸给药4周后,高剂量给药组可显著性降低糖尿病模型小鼠血糖、甘油三酯、总胆固醇、血肌酐和尿素水平;玉泉丸高剂量给药组可显著降低糖尿病模型小鼠肾脏指数;RT-PCR结果显示,玉泉丸可显著性下调microRNA-320基因表达,上调PI3K、Akt以及GLUT-1基因表达;Western Blot结果显示,玉泉丸可显著性上调PI3K、Akt以及GLUT-1蛋白表达。结论:玉泉丸可有效改善糖尿病模型小鼠糖代谢水平,其作用机制可能至少与下调肾脏组织中microRNA-320过表达、上调PI3K/Akt信号通路介导的GLUT-1过表达有关。Objective:To investigate the effect of Yuquan pill on glucose metabolism in diabetic mice by regulating microRNA-320 and elucidate its mechanism.Methods:Blood glucose,triglyceride,total cholesterol,serum creatinine,and urea were detected by biochemical analyzer.RT-PCR detected the mRNA expression of microRNA-320,PI3K,Akt and GLUT-1.Western Blot was used to detect PI3K,Akt and GLUT-1 protein expression.Results:After 4 weeks of Yuquan pill administration,high dose of Yuquan pill could significantly reduce blood glucose,triglyceride,total cholesterol,serum creatinine,and urea in diabetic model mice.High dose of the Yuquan pill can significantly reduce the kidney index of diabetic model mice.The results of RT-PCR showed that Yuquan pill could significantly down-regulate the expression of microRNA-320 gene,and up-regulate the expression of PI3K,Akt and GLUT-1 gene.Western Blot results showed that Yuquan pill could significantly up-regulate the expression of PI3K,Akt,and GLUT-1 protein.Conclusion:Yuquan pill can effectively improve glucose metabolism in diabetic mice by down-regulating microRNA-320 over-expression and up-regulating GLUT-1 over-expression mediated by PI3K/Akt signaling pathway in kidney tissue.
