肿节风提取物上调NKTR抑制小鼠肺癌生长实验研究

摘　　要：目的:探讨肿节风对肺癌的作用及其机制。方法:CCK8法检测肿节风对肺癌细胞LLC细胞活力的影响,LLC小鼠模型评价肿节风体内抗癌活性,蛋白质组学筛选肿节风处理的LLC细胞中的靶蛋白,蛋白质印迹实验验证靶蛋白的表达,流式细胞术检测小鼠外周血T细胞和NK细胞。结果:CCK8检测结果表明,肿节风抑制LLC细胞的增殖。肿节风治疗显著抑制LLC荷瘤小鼠的肿瘤生长。蛋白质组学分析表明,肿节风上调肺癌细胞中NKTR蛋白表达。Western Blot检测结果显示,肿节风干预的肺癌细胞及小鼠肿瘤组织中NKTR蛋白表达量升高。GEPIA数据库分析显示,NKTR在肺癌组织中表达量低于癌旁组织。肿节风治疗的荷瘤小鼠外周血NK细胞和T细胞比例显著升高。结论:肿节风通过上调NKTR表达增强抗肿瘤免疫。Objective:To investigate the effect of Sarcandra glabra on lung cancer and its mechanism.Methods:The CCK8 assay was used to detect the effect of Sarcandra glabra on the viability of LLC lung cancer cells.The LLC mouse model was used to evaluate the anti-cancer activity of Sarcandra glabra in vivo.Proteomics was employed to screen for target proteins in LLC cells treated with Sarcandra glabra,and Western Blot experiments were conducted to verify the expression of target proteins.Flow cytometry detected T cells and NK cells in mouse peripheral blood.Results:The CCK8 assay showed that Sarcandra glabra inhibited the proliferation of LLC cells.Sarcandra glabra treatment significantly suppressed tumor growth in LLC tumor-bearing mice.Proteomic analysis revealed that Sarcandra glabra upregulated the expression of NKTR protein in lung cancer cells,Western Blot assay showed elevated NKTR protein expression in Sarcandra glabra intervened in lung cancer cells and mouse tumor tissues.GEPIA database analysis showed that NKTR expression was lower in lung cancer tissues than in paracancerous tissues.The proportion of NK cells and T cells in the peripheral blood of tumor-bearing mice treated with Sarcandra glabra was significantly increased.Conclusion:Sarcandra glabra enhances antitumor immunity by upregulating NKTR expression.
