摘 要:目的:应用蛋白芯片技术筛选环磷酰胺(CTX)诱导的骨髓抑制小鼠模型的差异蛋白,从蛋白水平揭示艾炷灸改善CTX骨髓抑制机制。方法:将C57BL/6实验小鼠随机分为对照组、模型组和艾灸组,实验结束后检测各组小鼠外周血细胞数量,并从各组抽取3例血清样本,采用抗体蛋白芯片检测并分析3组间的差异蛋白。结果:艾灸组小鼠外周血WBC、Mon、Gra、PLT、RBC的数量高于对照组(P<0.05),蛋白芯片共筛选出差异蛋白12种,对照组与模型组间的差异蛋白为10种,分别是:ICAM-1、MIP-1g、BLC、TNF RI、TNF RII、PF4、IL-1a、IL-4、IL-10、IL-17;艾灸组与模型组间的差异蛋白有2种,分别是MIP-1g、TNF RI;最终得出共同差异蛋白为:MIP-1g、TNF RI。结论:艾炷灸改善CTX骨髓抑制模型的血清差异蛋白为MIP-1g、TNF RI,其起效机制可能是通过调节外周血细胞因子MIP-1g和TNF RI的表达实现造血干细胞的动员。Objective:This study aimed to identify differential proteins in a cyclophosphamide(CTX)-induced bone marrow suppression mouse model using protein chip technology,and to elucidate the mechanism by which moxibustion improves CTX-induced bone marrow suppression at the protein level.Methods:C57BL/6 mice were randomly divided into control,model,and moxibustion groups.After the experiment,peripheral blood cell counts were measured,and serum samples were collected from each group.Differential proteins among the three groups were detected and analyzed using antibody protein chips.Results:The numbers of peripheral blood WBC,Mon,Gra,PLT and RBC in the moxibustion group were higher than those in the control group(P<0.05).A total of 12 differential proteins were identified by protein chip analysis,with 10 differential proteins between the control and model groups,including ICAM-1,MIP-1g,BLC,TNF RI,TNF RII,PF4,IL-1a,IL-4,IL-10,and IL-17.There were 2 differential proteins between the moxibustion and model groups,namely MIP-1g and TNF RI.The common differential proteins identified were MIP-1g and TNF RI.Conclusion:Moxibustion improves serum differential proteins in the CTX-induced bone marrow suppression model,namely MIP-1g and TNF RI.The mechanism of action may involve regulating the expression of peripheral blood cytokines MIP-1g and TNF RI to mobilize hematopoietic stem cells.
