益景汤对糖尿病大鼠视网膜Occludin、CⅣ及PDGF-B的影响

摘　　要：目的:探讨益景汤对糖尿病(DM)大鼠视网膜微血管的保护效应及机制。方法:18只大鼠被随机分为3组:正常对照组、糖尿病对照组(DM对照组)、益景汤治疗组,每组6只。采用链脲佐菌素对DM对照组、益景汤治疗组大鼠进行DM造模。正常对照组和DM对照组采用生理盐水进行灌胃干预,益景汤治疗组采用益景汤灌胃干预。12周后处死所有大鼠,RT-PCR法检测Occludin mRNA、CⅣmRNA、PDGF-B mRNA的表达量,Western Blot法检测视网膜组织结构蛋白Occludin、Ⅳ型胶原蛋白表达。结果:视网膜Occludin、CⅣ及PDGF-B mRNA的表达:DM对照组大鼠视网膜中Occludin mRNA表达量降低,CⅣ及PDGF-B mRNA表达量上升,与正常组相比,差异均具有统计学意义(均P<0.05);益景汤能抑制DM大鼠视网膜Occludin mRNA表达量降低,抑制CⅣmRNA及PDGF-B mRNA表达量上升,益景汤治疗组上述mRNA的表达量与DM对照组相比,差异具有统计学意义(均P<0.05)。视网膜Occludin、CⅣ蛋白表达:DM对照组Occludin蛋白表达量下调、CⅣ蛋白表达量上升,与正常组相比差异均具有统计学意义(均P<0.05);益景汤能抑制DM大鼠视网膜Occludin蛋白表达量的下降、CⅣ蛋白表达量的上升,益景汤治疗组上述蛋白的表达量与DM组相比差异具有统计学意义(均P<0.05)。结论:益景汤能抑制DM大鼠视网膜Occludin下降、调控结构蛋白CⅣ上升、抑制PDGF-B表达,保护微血管完整性,降低BRB的损害。Objective:To investigate the protective effect and mechanism of Yijing Decoction on retinal microvessels in diabetic(DM)rats.Methods:Eighteen rats were randomly divided into 3 groups:normal control group,diabetes control group(DM control group),Yijing decoction treatment group,with 6 rats in each group.DM modeling was performed on the rats in DM control group and Yijing decoction treatment group with streptozotocin.Normal control group and DM control group were treated with normal saline and Yijing Decoction was treated with Yijing decoction.After 12 weeks,all rats were killed.RT-PCR was used to detect the expression of Occludin mRNA,CⅣmRNA and PDGF-B mRNA,and Western blot was used to detect the expression of retinal structural proteins occludin and typeⅣcollagen.Results:Expression of retinal Occludin,CⅣand PDGF-B mRNA:Compared with normal group,the mRNA expression of retinal occludin in DM control group decreased,while the mRNA expression of CⅣand PDGF-B increased,with statistical significance(all P<0.05).Yijing Decoction could inhibit the decrease of retinal occludin mRNA expression and the increase of CⅣmRNA and PDGF-B mRNA expression in DM rats,and the above mRNA expression in Yijing Decoction group had statistical significance compared with DM control group(all P<0.05).Expression of retinal occludin and CⅣprotein:The expression of occludin protein in the DM control group was down-regulated and the protein expression of CⅣwas increased,which were statistically different from those in the normal group(all P<0.05).Yijing Decoction could inhibit the decrease of retinal occludin protein expression and the increase of CⅣprotein expression in DM rats,and the above protein expression in Yijing Decoction group had statistical significance compared with DM group(all P<0.05).Conclusion:Yijing Decoction can protect microvascular integrity and reduce BRB damage by inhibiting retinal occludin decline,structural protein CⅣrise and PDGF-B expression in DM rats.
