摘 要:目的:利用GEO数据库及网络药理学研究半枝莲、白花蛇舌草治疗乳腺癌肺转移和骨转移分子机制。方法:采用R语言差异分析GEO芯片获取疾病靶点;检索TCMSP、PubChem、SwissTargetPrediction数据库筛选药物有效成分的作用靶点;将药物靶点与疾病靶点取交集,通过STRING数据库构建PPI网络,并用Cytoscape进行可视化和拓扑分析,最后利用Web Gestalt数据库进行GO、KEGG富集分析。结果:经筛选后半枝莲主要包括28个有效成分,如黄芩素、木犀草素、槲皮素等,白花蛇舌草包括7个有效成分,如槲皮素、2,3-二甲氧基-6-甲基蒽醌等,半枝莲靶基因504个,白花蛇舌草靶基因313个。GEO数据库中分别筛选到364、4674个乳腺癌肺转移、骨转移差异基因。PPI网络各筛选出10个核心靶点。GO和KEGG富集分析表明,半枝莲、白花蛇舌草可通过影响转录调节复合物、转移酶复合物等细胞组分,调节β-catenin聚集等分子功能,干预Wnt、细胞周期、TGF-β等信号通路,参与调控细胞周期、T细胞活化等生物进程,治疗乳腺癌肺转移、骨转移则可能通过影响蛋白激酶复合物等细胞组分,调节类固醇结合、氧化还原酶活性等分子功能,干预细胞周期、P53、PI3K/Akt、FoxO等信号通路,参与类固醇代谢等进程。结论:初步揭示半枝莲、白花蛇舌草抗乳腺癌肺转移、骨转移的作用靶标和潜在机制,发现其作用是多成分、多靶点、多途径的,为今后分子生物学实验奠定基础。Objective:To investigate The molecular mechanism of Sculellaria barbata and Hedyotis diffusa in the treatment of breast cancer with lung metastasis and bone metastasis was studied by GEO database and network pharmacology.Methods:The disease target was obtained by using R language difference analysis of GEO chip;TCMSP,PubChem and SwissTargetPrediction database were searched to screen the action targets of drug active ingredients;The intersection of drug targets and disease targets was used to construct PPI network through STRING database,Visualization and topological analysis were performed using Cytoscape;Finally,the Web Gestalt database was used for GO and KEGG enrichment analysis.Results:The results showed that there were 28 effective components in Sculellaria barbata,such as baicalin,luteolin,quercetin and so on.Hedyotis diffusa includes 7,such as quercetin,2,3-dimethoxy-6-methylanthraquinone and so on.The target genes of Sculellaria barbata were 504,The target genes of Hedyotis diffusa were 313.In GEO database,364 and 4674 differential genes for breast cancer lung metastasis and bone metastasis were screened.PPI network screened 10 core targets.GO and KEGG analysis showed that the drugs could affect the transcriptional regulatory complex,transferase complex and other cellular components,regulate the molecular functions such asβ-catenin aggregation,interfere with Wnt,cell cycle,TGF-βand other signaling pathways,participate in the regulation of cell cycle,T cell activation and other life processes,and treat breast cancer lung metastasis;The treatment of breast cancer bone metastasis may affect the protein kinase complex and other cell components,regulate steroid binding,redox enzyme activity and other molecular functions,interfere with cell cycle,P53,PI3K/Akt,FoxO and other signaling pathways,and participate in steroid metabolism and other life processes.Conclusion:This study preliminarily revealed the target and potential mechanism of Sculellaria barbata and Hedyotis diffusa therapy for lung metastasis and b
