摘 要:为探究注射褪黑素对急性铅中毒小鼠卵母细胞体外成熟的影响,以6~8周龄体况相近的ICR小鼠卵母细胞为研究对象,腹腔注射不同剂量的褪黑素(20、30、40 mg/kg体重),以第一极体排出作为卵母细胞成熟的标志,筛选出褪黑素适宜注射剂量继续进行后续试验。通过活性氧水平、ATP含量、线粒体分布以及细胞凋亡检测,分析注射褪黑素对急性铅中毒小鼠卵母细胞体外成熟的影响。结果显示,腹腔注射40 mg/kg的褪黑素可显著降低卵母细胞内活性氧水平、卵母细胞线粒体异常分布比率以及早期卵母细胞凋亡率,显著提高卵母细胞第一极体排出率及ATP含量。结果表明,腹腔注射40 mg/kg褪黑素可以提高急性铅中毒小鼠卵母细胞体外成熟的效果。In order to investigate the effect of in vivo injection of melatonin on the maturation of oocytes in mice with acute lead poisoning,the oocytes of ICR mice aged 6—8 weeks with similar body condition were intraperitoneally injected with different doses of melatonin(20,30,40 mg/kg weight) in vivo,and the release of the first polar body was used as a sign of oocyte maturation.The optimal injection concentration of melatonin was determined,and the impact of melatonin injection on the in vitro maturation of mice oocytes with acute lead poisoning was analyzed by assessing reactive oxygen species,ATP content,mitochondrial distribution and apoptosis.The results showed that intraperitoneal injection of 40 mg/kg melatonin significantly reduced the reactive oxygen species in oocytes,the abnormal distribution ratio of mitochondria,and the early apoptosis rate of oocytes.Additionally,it and significantly increased the first polar body excretion rate and ATP content of oocytes.In conclusion,intraperitoneal injection of 40 mg/kg melatonin enhances the effect of oocyte maturation in vitro in mice with acute lead poisoning.
