摘 要:C3H转录因子属于锌指蛋白(zinc finger protein,ZFP)家族,在植物应对生物和非生物胁迫中发挥重要作用。为了探究C3H基因在蒺藜苜蓿(Medicago truncatula)中的潜在功能,本研究从蒺藜苜蓿R108中克隆MtC3H基因(GenBank No.NC_053045.1)全长序列,该基因CDS全长672 bp,编码223个氨基酸。进化树分析显示MtC3H蛋白与鹰嘴豆(Cicer arietinum)和豌豆(Pisum sativum)亲缘关系较近。表达模式分析表明,MtC3H在蒺藜苜蓿茎中的表达量最高,根次之;该基因能够响应外源激素赤霉素(gibberellin,GA)、生长素(indole acetic acid,IAA)和茉莉酸甲酯(methyl jasmonate,MeJA)的诱导,且对MeJA的诱导更敏感。亚细胞定位分析显示,该蛋白定位在细胞核和细胞质。酵母表达分析表明,MtC3H蛋白不具有毒性,存在转录自激活活性。本研究为深入探究MtC3H基因的作用提供了参考。C3H transcription factors belong to the zinc finger protein(ZFP)family and play a crucial role in plant responses to both biotic and abiotic stresses.To investigate the potential function of the MtC3H gene in Medicago truncatula,the full-length sequence of the MtC3H gene was cloned from Medicago truncatula R108.the full length of CDS of MtC3H was 672 bp and encoded 223 amino acids.Phylogenetic tree analysis showed that MtC3H protein was closely related to Cicer arietinum and Pisum sativum.Expression pattern analysis demonstrated that MtC3H exhibited the highest expression level in the stems of Medicago truncatula,followed by the roots.Furthermore,the MtC3H gene was found to respond to exogenous hormones such as gibberellin(GA),auxin(IAA),and methyl jasmonate(MeJA),with a particularly heightened sensitivity to MeJA induction.Subcellular localization analysis indicated that the MtC3H protein predominantly localized in the nucleus and cytoplasm.Notably,yeast expression analysis revealed that the MtC3H protein exhibited nontoxic properties and possessed transcriptional self-activation activity.This study provides a reference for further exploring the role of MtC3H gene.
