摘  要:蔗糖合成酶(SuSy)是植物进行蔗糖代谢的关键酶之一,控制着植物体内糖分的合成和运输,在植物的生长和代谢中起着重要作用。作为唯一的催化蔗糖分解与合成的可逆反应酶,蔗糖合成酶及其功能一直是揭示植物生长发育规律的重要研究内容。本研究以生物信息学手段对慈姑SuSy基因家族及其成员进行鉴定和表达分析。结果表明,慈姑SuSy家族共获得83个SuSy基因。这些基因编码的蛋白质的氨基酸长度位于180~1084 aa之间,等电点变化位于4.52~9.56之间,平均值为6.29,大部分蛋白表现呈酸性。与马铃薯和拟南芥进化比较,110个SuSy基因被分成11个亚族,马铃薯和拟南芥中的SuSy基因全部存在于Ⅰ、Ⅱ、Ⅳ组中。同时,热图分析发现83个SuSy基因成员在2个慈姑品种中的表达存在明显差异,且不同的球茎发育阶段也存在差异表达,说明它们在不同品种慈姑中的功能存在差异。本研究为后续对慈姑SuSy基因功能研究及蔗糖-淀粉代谢调控的应用提供理论基础。Sucrose synthase(SuSy)is one of the key enzymes for sucrose metabolism in plants,which controls the synthesis and transportation of sugar in plants and plays an important role in the growth and metabolism of plants.As the only reversible enzyme that catalyzes the decomposition and synthesis of sucrose,sucrose synthase and its function have always been an important research content to reveal the laws of plant growth and development.In this study,the SuSy gene family of Sagittaria sagittifolia and its members were identified and expressed by bioinformatics.The result showed that 83 SuSy genes were obtained from SuSy family of Sagittaria sagittifolia.The proteins encoded by these genes have amino acid lengths ranging from 180 to 1084,and the isoelectric point varied from 4.52 to 9.56,with an average value of 6.29.Most proteins were acidic.Compared with potato and Arabidopsis,110 SuSy genes were divided into 11 subgroups,and all SuSy genes in potato and Arabidopsis existed in groupsⅠ,ⅡandⅣ.At the same time,heat map analysis showed that the expression of SuSy gene members in two varieties of Sagittaria sagittifolia was significantly different,and there were also different expressions in different bulb development stages,which indicated that their functions in different varieties of Sagittaria sagittifolia were different.This study provides a theoretical basis for further research on the function of Sagittaria sagittifolia gene about SuSy and the application of sucrose-starch metabolism regulation.
