摘 要:为明确玫瑰抗虫防御酶活性及空间传导特性,本研究采用机械损伤方式处理玫瑰叶片,观测防御酶活性在空间与时间二维度的变化情况。结果表明:玫瑰部分叶片被针刺处理后,不同部位POD、PPO、PAL活性随时间发生明显变化,但防御酶对机械损伤的应答反应并不相同。处理后不同部位POD、PPO活性均升高,变化趋势一致,均在48 h时达到最高峰,PPO活性在48、96 h时显著高于CK(P<0.01),POD活性在48 h时显著高于CK(P<0.01),未损伤部位升高较明显。PAL活性变化与POD、PPO较为不同,损伤部位PAL活性迅速升高,15~96 h时显著高于其他2组,未损伤部位被激活较为迟缓,在168~240 h时显著高于CK(P<0.05)。处理后防御酶在空间上的变化是有差异的,中、下层POD活性随时间呈一过性增加状态,随后降低并回到原初活性状态,拐点呈“下层>中层>上层”顺序,各层PAL活性增加呈现“中、下、上”时效先后特征,而PPO活性空间各层时效一致,均为先上升后下降,不存在明显的先后差异。本研究采用了机械损伤诱导玫瑰产生抗虫防御,得到了损伤与未损伤部位的应激反应及在空间传导协同性的理论数据。在实际生产中可选择在采收前7~10 d选取中层少量叶片进行针刺,提高玫瑰抗虫防御以减少虫害,为降低化药使用,虫害的物理防控提供了一种重要手段。In order to clarify the anti-pest defense enzyme activity and spatial conduction characteristics of roses,this study treated rose leaves with mechanical damage to observe activities of anti-pest defense enzymes in space and time.The results showed that the activities of POD,PPO and PAL in different parts of rose leaves were significantly changed with time,but the responses of defense enzymes to mechanical injury were different.After treatment,the activities of POD and PPO in different parts increased,with the same trend,and reached the peak at 48 h.The activity of PPO at 48 and 96 h was significantly higher than CK(P<0.01),and the activity of POD at 48 h was significantly higher than CK(P<0.01),and the increase of undamaged parts was more obvious.The change of PAL activity was different from that of POD and PPO,PAL activity increased rapidly in the injury and was significantly higher than that in the other two groups at 15 to 96 h.The activation of the undamaged site was slower,and was significantly higher than CK at 168 to 240 h(P<0.05).After processing defense enzymes in space there is a difference,the change of middle and lower POD activity increases with time in a transient state,then reduce the activity and return to original state,inflection point is"lower layer>middle layer>upper layer"order,the increase of PAL activity in each layer showed the aging sequence of"middle,lower and upper",while the aging sequence of PPO activity space was the same,which increased first and then decreased,and there was no significant difference in the order.In this study,mechanical damage induced anti-insect defense was used to study the stress response and spatial conduction characteristics of various parts.In actual production,a small number of leaves in the middle layer can be selected for needling 7 to 10 days before harvest to improve insect defense and reduce insect pests.It provided an important means for reducing the use of chemical drugs and pest control.
