摘 要:乌药(Lindera aggregata)属于一种樟科植物,其干燥块根是传统中药,具有抗氧化、抗炎、镇痛等多种药理活性。本研究探讨了乌药醇提取物联合耐力训练对NAFLD大鼠的干预效果及其通过SIRT1介导的线粒体功能改善机制。实验将大鼠分为对照组、模型组、乌药醇提取物不同剂量组、耐力训练组和联合治疗组。结果显示,模型组大鼠血清ALT、AST、TC、TG和LDL-c水平显著升高,肝脏脂质积累明显,线粒体结构受损,并伴有氧化应激和炎症反应。乌药醇提取物和耐力训练均能显著降低上述生化指标和病理改变,且联合治疗效果更为显著。联合治疗组显著上调了SIRT1、PPAR-α和mCPT-1的表达水平,下调了CD36的表达水平,改善了肝脏线粒体功能,减轻了氧化应激和炎症反应。本研究表明,乌药醇提取物联合耐力训练在改善NAFLD大鼠肝脏脂代谢、氧化应激和炎症反应方面具有协同作用,且通过SIRT1信号通路改善线粒体功能,为NAFLD的治疗提供了新的思路和方法。Lindera aggregata,a plant in the Lauraceae family,is a traditional Chinese medicine with dried roots that exhibit various pharmacological activities,including antioxidant,anti-inflammatory,and analgesic properties.This study investigates the effects of Lindera aggregata extract combined with endurance training on rats with non-alcoholic fatty liver disease(NAFLD)and the mechanism of improvement through SIRT1-mediated mitochondrial function.Rats were divided into control,model,various doses of Lindera aggregata extract,endurance training,and combination treatment groups.Results showed that in the model group,serum levels of ALT,AST,TC,TG,and LDL-c were significantly elevated,hepatic lipid accumulation was evident,mitochondrial structure was damaged,and oxidative stress and inflammation were present.Both Lindera aggregata extract and endurance training significantly reduced these biochemical and pathological changes,with the combination treatment showing more pronounced e ffects.The combination treatment group significantly upregulated the expression of SIRT1,PPAR-α,and mCPT-1,while downregulating CD36 expression,improving hepatic mitochondrial function,and reducing oxidative stress and inflammation.This study demonstrates that Lindera aggregata extract combined with endurance training has a synergistic effect on improving hepatic lipid metabolism,oxidative stress,and inflammation in NAFLD rats,and improves mitochondrial function through the SIRT1 signaling pathway,providing new insights and methods for the treatment of NAFLD.
