摘  要:为实现动物性食品中氟喹诺酮类药物(FQ)残留量的高通量检测,合成与FQ有特异性响应的荧光N-掺杂碳点(N-CDs);以柠檬酸为碳源,L-酪氨酸为氮源,通过一步水热法制备N-CDs。利用透射电子显微镜、紫外-可见分光光度计、荧光分光光度计等仪器对N-CDs的结构、组成和光学性质进行了研究。基于FQ对N-CDs的选择性荧光增强作用,建立FQ的含量检测方法,并将其应用于鸡肉中洛美沙星的含量分析。所制备的N-CDs的平均粒径为18.25 nm,表面有羟基、羧基、酰胺基等亲水官能团,具有良好的水溶性、耐盐性、稳定性,荧光量子产率为3.63%。测定FQ的线性范围为0.781~400μg/mL,检测限在0.114~0.582μg/mL之间(S/N=3)。用于市售鸡肉中洛美沙星的检测,加标回收率为98.59%~104.3%,RSD在0.34%~5.46%之间。该实验所建立的检测方法为动物性食品中FQ的残留检测提供一种快速、灵敏的高通量检测方法。In order to achieve high-throughput detection of fluoroquinolones(FQ)residues in animal foods,fluorescent N-doped carbon dots(N-CDs)with specific response to FQ were synthesized.N-CDs were prepared by one-step hydrothermal method using citric acid as carbon source and L-tyrosine as nitrogen source.The structure,composition and optical properties of N-CDs were studied by transmission electron microscopy,ultraviolet-visible spectrophotometer,fluorescence spectrophotometer,etc.Based on the selective fluorescence enhancement effect of FQ on N-CDs,a FQ content detection method was established and applied to the content analysis of Lomefloxacin in chicken.The average particle size of the prepared N-CDs was 18.25 nm,and the surface contained hydrophilic functional groups such as hydroxyl,carboxyl and amide groups.It had good water solubility,salt tolerance and stability,and the fluorescence quantum yield was 3.63%.The linear range of FQ was 0.781-400μg/mL,and the detection limit was 0.114-0.582μg/mL(S/N=3).The recovery rate was 98.59%-104.3%and RSD was 0.34%-5.46%for the detection of lomefloxacin in commercially available chicken.The detection method established in this experiment provides a rapid and sensitive high-throughput detection method for the detection of FQ residues in animal food.
