摘  要:目的:建立磁场辅助肝脏冷保存平台,探究静磁场对冷保存肝脏的保护作用。方法:通过检测保存后各组SD大鼠肝组织中活性氧(ROS)、丙二醛(MDA)、和超氧化物歧化酶(SOD)水平评估各组肝脏氧化应激水平差异。通过苏木精-伊红(HE)染色对比各组肝脏组织病理学变化。通过干湿重比对比各组肝脏保存后水肿情况,各组间采用单因素方差分析来进行统计分析。结果:静磁场辅助肝脏保存平台可以产生稳定的磁场;20~50 Gs磁场辅助静态冷保存(SCS)组较单纯SCS组肝脏组织形态更完整;ROS[303.23±21.49比333.63±14.04, P>0.05;250.3±21.48比333.63±14.04, P<0.01;286.84±30.43比333.63±14.04, P<0.05;295.99±26.69比333.63±14.04, P>0.05;F(4,20)=8.204]、MDA[4.98±1.57比6.71±0.84, P<0.01;4.13±0.40比6.71±0.84, P<0.01;4.98±0.99比6.71±0.84, P<0.01;5.18±1.24比6.71±0.84, P<0.05;F(4,20)=10.090]水平低于对照组,SOD[5.72±1.48比4.45±0.84, P>0.05;8.02±1.66比4.45±0.84, P<0.01;6.53±0.56比4.45±0.84, P>0.05;6.28±1.21比4.45±0.84, P>0.05;F(4,20)=5.675]水平高于对照组。磁场组干湿重比优于对照组[0.31±0.03比0.25±0.04, P<0.05;F(2,12)=112.500]。 结论:成功构建静磁场辅助肝脏冷保存平台,其可在多个层面提高肝脏在冷藏过程中的抗氧化能力,从而提高肝脏保存质量。Objective:The aim was to establish a liver cold preservation platform aided by a magnetic field and to investigate the protective effects of static magnetic fields on cold-preserved livers.Methods:The team assessed oxidative stress variations among different groups by measuring reactive oxygen(ROS),malondialdehyde(MDA),and superoxide dismutase(SOD)levels in the liver tissues post-preservation.They compared histological alterations using hematoxylin eosin(HE)staining.One-way ANOVA was used for statistical analysis among all groups.Results:A stable magnetic field is produced by the static magnetic field-assisted liver preservation platform;liver tissue integrity is better preserved in the group assisted by a magnetic field compared to the group preserved with SCS alone;moreover,ROS[303.23±21.49 vs.333.63±14.04,P>0.05;250.3±21.48 vs.333.63±14.04,P<0.01;286.84±30.43 vs.333.63±14.04,P<0.05;295.99±26.69 vs.333.63±14.04,P>0.05;F(4,20)=8.204],MDA[4.98±1.57 vs.6.71±0.84,P<0.01;4.13±0.40 vs.6.71±0.84,P<0.01;4.98±0.99 vs.6.71±0.84,P<0.01;5.18±1.24 vs.6.71±0.84,P<0.05;F(4,20)=10.090]levels were reduced relative to control group and the level of SOD was higher than that of control group[5.72±1.48 vs.4.45±0.84,P>0.05;8.02±1.66 vs.4.45±0.84,P<0.01;6.53±0.56 vs.4.45±0.84,P>0.05;6.28±1.21 vs.4.45±0.84,P>0.05;F(4,20)=5.675].The dry to wet weight ratio was better in the magnetic field group than in the control group[0.31±0.03 vs.0.25±0.04,P<0.05;F(2,12)=112.500].Conclusion:The development of a static magnetic field-assisted liver cold storage platform significantly enhances the liver’s resistance to oxidation during storage,improving preservation quality.This research offers innovative approaches for enhancing liver cold storage methods and is expected to serve as fundamental groundwork for critical medical practices,such as liver transplantation,in the future.
