骨软骨前体细胞与骨髓间充质干细胞相关特性的比较研究

摘  要:目的:从晚期骨关节炎患者关节软骨和骨髓组织中分离出CD146+骨软骨前体细胞(CD146+OCPs)和骨髓间充质干细胞(BMSCs),检测并比较CD146+OCPs与BMSCs的相关特性。方法:收集5例在武汉市第四医院行人工全膝关节置换术的晚期骨关节炎患者术中废弃的软骨组织和骨髓,分离培养出CD146+OCPs与BMSCs,培养3代后,分别用流式细胞仪检测其表面抗原表达,比较其克隆形成及成软骨、成骨、成脂分化能力。运用独立样本t检验进行组间比较,计数资料则采用χ^(2)检验。结果:CD146+OCPs高表达CD29、CD44、CD73及CD90等表面抗原分子,而几乎不表达CD34、CD45及CD133等抗原分子,这与BMSCs表面抗原表达谱相似;CD146+OCPs克隆效率为(20.8±3.8)%,BMSCs克隆效率为(19.3±3.1)%,两组差异无统计学意义(t=0.749,P>0.05)。与BMSCs比较,CD146+OCPs成骨分化能力较强,而成脂分化能力较弱,成软骨分化能力并无统计学意义。结论:CD146+OCPs与BMSCs形态功能类似。Objective:To isolate CD146+osteochondroprogenitors(OCPs)and bone marrow mesenchymal stem cells(BMSCs)from joint cartilage and bone marrow tissues of patients with advanced osteoarthritis,and to detect and compare the related characteristics of CD146+OCPs and BMSCs.Methods:Collected discarded cartilage tissue and bone marrow from patients with advanced osteoarthritis who underwent total knee arthroplasty in Wuhan Forth Hospital.Separated and cultured CD146+OCPs and BMSCs.After 3 generations of cultivation,detected their surface antigen expression using flow cytometry,and compared their clone formation,chondrogenic,osteogenic,and adipogenic differentiation abilities.Independent sample t-test was used for comparison between the two groups,while chi-square test was used for count data.Results:CD146+OCPs highly expressed surface antigen molecules such as CD29,CD44,CD73,and CD90,while almost no antigen molecules such as CD34,CD45,and CD133 were expressed,which was similar to the surface antigen expression profile of BMSCs;The cloning efficiency of CD146+OCPs was(20.8±3.8)%,and the cloning efficiency of BMSCs was(19.3±3.1)%,with no statistically significant difference between the two groups(t=0.749,P>0.05).Compared with BMSCs,CD146+OCPs had stronger osteogenic differentiation ability,weaker adipogenic differentiation ability,and no significant difference in chondrogenic differentiation ability.Conclusion:CD146+OCPs had similar morphology and function to BMSCs.
