摘  要:目的:探讨脊髓损伤大鼠铜死亡相关蛋白(FDX1、LIAS、LIPT1、DLD、DLAT、PDHA1、PDHB、MTF1、GLS、CDKN2A、ATP7B、SLC31A1)的表达以及与脊髓功能的相关性。方法:20只小鼠随机分为对照组和脊髓损伤组,每组10只,对照组小鼠接受无脊髓损伤手术,脊髓损伤组小鼠接受挫伤法建立脊髓损伤模型。建模1周后采用BMS(Basso mouse scale)评分和脊髓损伤运动功能(BBB)评分对两组小鼠神经功能恢复和运动功能评分;采用蛋白质免疫印迹分析两组小鼠脊髓组织铜死亡相关蛋白[FDX1、硫辛酰合成酶(LIAS)、脂酰转移酶1(LIPT1)、二氢硫辛酸脱氢酶(DLD)、二氢硫辛酰胺乙酰转移酶(DLAT)、丙酮酸脱氢酶E1亚基α1(PDHA1)、丙酮酸脱氢酶E1亚基β(PDHB)、金属调节转录因子1(MTF1)、谷氨酰胺酶(GLS)、细胞周期蛋白依赖性激酶抑制蛋白2A(CDKN2A)、ATP7B、SLC31A1]的表达水平;采用试剂盒测定铜离子水平;采用尼氏染色计算神经元数量。组间计量数据比较采用独立样本t检验。结果:对照组细胞铁死亡相关蛋白FDX1、LIAS、LIPT1、DLD、DLAT、PDHA1、PDHB、ATP7B、SLC31A1水平(0.98±0.08、0.99±0.07、1.01±0.11、0.99±0.11、0.83±0.13、0.85±0.09、0.94±0.12、0.84±0.09、1.05±0.14)明显低于脊髓损伤组小鼠(1.49±0.10、1.51±0.11、1.55±0.14、1.45±0.13、1.50±0.19、1.50±0.16、1.55±0.12、1.27±0.12、1.55±0.14),差异有统计学意义(t=12.770、12.720、9.548、8.566、9.142、10.950、11.190、9.062、10.330,P<0.05)。对照组细胞铁死亡相关蛋白MTF1、CDKN2A、GLS水平(1.04±0.16、0.89±0.12、0.97±0.10)明显高于脊髓损伤组小鼠(0.69±0.11、0.58±0.07、0.65±0.07),差异有统计学意义(t=5.509、6.684、8.032,P<0.05)。对照组脊髓组织铜离子水平[(21.87±2.16)μmol/L]明显低于脊髓损伤组小鼠[(37.31±4.92)μmol/L],差异有统计学意义(t=9.108,P<0.05)。对照组小鼠BBB评分和BMS评分[(21.00±0.00)、(9.00±0.00)分]明显高于脊髓损伤组小�Objective:To investigate the expression of copper death related proteins(FDX1,LIAS,LIPT1,DLD,DLAT,PDHA1,PDHB,MTF1,GLS,CDKN2A,ATP7B,SLC31A1)and their correlation with spinal cord function in rats with spinal cord injury.Methods:20 mice were randomly divided into control group and spinal cord injury group(10 mice/per group).Mice in sham operation group received operation without spinal cord injury,and mice in spinal cord injury group received contusion method to establish spinal cord injury model.After 1 week of modeling,Basso mouse scale(BMS)score and Basso,Beattie and Bresnahan(BBB)score were used to evaluate the neural function recovery and motor function of the two groups of mice.The expression levels of copper death related proteins(FDX1,LIAS,LIPT1,DLD,DLAT,PDHA1,PDHB,MTF1,GLS,CDKN2A,ATP7B,SLC31A1)in the spinal cord of two groups of mice were analyzed by Western blotting.The copper ion level was determined by the kit.The number of neurons was calculated by Nissl staining.Independent sample T test was used to compare measurement data between groups.The measurement data between groups were compared by independent sample t test.Results:The levels of iron death related proteins FDX1,LIAS,LIPT1,DLD,DLAT,PDHA1,PDHB,ATP7B and SLC31A1 in control group(0.98±0.08,0.99±0.07,1.01±0.11,0.99±0.11,0.83±0.13,0.85±0.09,0.94±0.12,0.84±0.09,1.05±0.14 was significantly lower than that in the spinal cord injury group(1.49±0.10,1.51±0.11,1.55±0.14,1.45±0.13,1.50±0.19,1.50±0.16,1.55±0.12,1.27±0.12,1.55±0.14,t=12.770,12.720,9.548,8.566,9.142,10.950,11.190,9.062,10.330,P<0.05).The levels of iron death related proteins MTF1,CDKN2A and GLS in control group(1.04±0.16,0.89±0.12,0.97±0.10)was significantly higher than that in spinal cord injury group(0.69±0.11,0.58±0.07,0.65±0.07,t=5.509,6.684,8.032,P<0.05).The level of copper ion in the spinal cord tissue of the control group[(21.87±2.16)μmol/L]was significantly lower than that of the spinal cord injury group[(37.31±4.92)μmol/L,t=9.108,P<0.05].The BBB score and
