摘 要:目的探讨采用Metafer5软件扫描辅助阅片用于淋巴细胞微核检测的准确度。方法从1403名放射工作人员中随机选取100人的外周血淋巴细胞微核玻片作为研究对象。采用Metafer5软件扫描辅助阅片后,将除主核外还含有其他物质(微核、核芽和杂质等)的细胞在油镜下确认核实,并以此作为人工验证结果。计算软件扫描辅助阅片的灵敏度、特异度和约登指数,并绘制ROC曲线,计算ROC曲线下面积。结果Metafer5软件扫描辅助阅片计数微核细胞、微核和微核率异常的灵敏度分别为90.1%、89.7%和100%,特异度分别为97.4%、97.2%和98.8%,约登指数分别为0.87、0.87和0.99。三者的ROC曲线下面积分别为0.94、0.94和0.99。结论Metafer5软件扫描辅助阅片能准确检测外周血淋巴细胞微核数,可用于大样本的放射工作人员微核检测。Objective To investigate the accuracy of the Metafer5 software scanning-assisted slide reading method for micronucleus detection in the radiological staff.Methods A total of 100 peripheral blood lymphocyte micronucleus slides were randomly selected from 1403 radiological workers.The Metafer5 software scanning-assisted slide reading method was used to read the slides.Then the cells containing other substances(micronuclei,nuclear buds,and impurities,etc.)besides the main nucleus were manually confirmed under the oil microscope and these results were used as manual verification results.Sensitivity,specificity,Youden index,and the area under ROC curves were used to evaluate the accuracy of the Metafer5 software scanning-assisted slide reading method.Results The sensitivity of Metafer5 software scanning-assi stedslide reading method for identifying micronucleuscells,micronucleus,and abnormal micronucleus rate were 90.1%,89.7%and 100%,respectively,and the specificity was 97.4%,97.2%,and 98.8%,respectively,and the Youden index was 0.87,0.87,and 0.99,respectively.The area under the ROC curve for these three indexes mentioned above were 0.94,0.94,and 0.99,respectively.Conclusion The Metafer5 software scanning-assisted slide reading method can accurately detect peripheral blood lymphocyte micronuclei,and it is feasible for micronucleus assay in large samples of radiological staff.
