摘 要:刚地弓形虫是一种专性细胞内寄生的人畜共患寄生虫,可感染人、野鼠及其他温血动物。为丰富我国野鼠弓形虫基因型的相关基础数据,对采自我国5个地区(浙江省、黑龙江省、内蒙古自治区、广西壮族自治区和山西省)的20份野鼠脑组织弓形虫阳性DNA样本进行基因型鉴定。通过靶向扩增弓形虫的10个基因位点(SAG1,5′-SAG2,3′-SAG2,alter.SAG2,BTUB,c22-8,c29-2,L358,PK1,Apico),鉴定野鼠感染的弓形虫基因型。结果表明:在20份阳性样品中,4份样品扩增出10个基因位点,3份样品扩增出9个位点,3份样品扩增出8个位点,1份样品扩增出7个位点。其中,浙江省和内蒙古自治区的样本分别检测出1个新基因型,广西壮族自治区的样本检测出5个新基因型和2个已知基因型(Toxo DB#94和Toxo DB#92),山西省的样本检测出2个新基因型。发现了Toxo DB#92,Toxo DB#94和9种可感染野鼠的新基因型,其中Toxo DB#92和Toxo DB#94首次在广西小家鼠体内发现。对我国不同地区野鼠弓形虫基因型进行鉴定,扩展了野鼠弓形虫基因型分布,以期为弓形虫病的研究提供参考。Toxoplasma gondii is an obligate intracellular zoonotic parasite that infects humans,wild mice and other warm-blooded animals.The aim of this study was to enrich the basic data of Toxoplasma gondii genotypes in China.In this study,genotypes of toxoplasma gondii positive DNA samples from 20 wild mice brain tissues of wild rodents from 5 regions of China(Zhejiang Province,Heilongjiang Province,Inner Mongolia Autonomous Region,Guangxi Zhuang Autonomous Region and Shanxi Province)were identified.Genotypes of toxoplasma infection in wild mice were identified by targeting 10 loci(SAG1,5′-SAG2,3′-SAG2,alter.SAG2,BTUB,c22-8,c29-2,L358,PK1,Apico)of T.gondii.The results showed that among of the 20 positive samples,4 samples were successfully amplified ten genetic loci,3 samples were amplified 9 genetic loci,3 samples amplified 8genetic loci and 1 sample amplified 7 genetic loci.One new genotype was detected in Zhejiang Province.A new genotype was detected in Inner Mongolia Autonomous Region.Five new genotypes and two known genotypes were detected in Guangxi Zhuang Autonomous Region,Toxo DB#94and Toxo DB#92.Two new genotypes were detected in Shanxi Province.This study reported genotypes of T.gondii in wild mice in different regions of China,extending the distribution of genotypes of T.gondii in wild mice in China,and providing basic data for the study of toxoplasmosis.
