摘 要:目的改进《中华人民共和国药典》黄连上清片项下盐酸小檗碱含量的测定方法。方法改进后的方法采用高效液相色谱法,以Welch Welchrom C18柱(4.6 mm×250 mm,5μm)为固定相;以乙腈-0.033 mol/L磷酸二氢钾(24∶76)为流动相,流速1.0 ml/min,检测波长345.0 nm。结果改进后,样品在此色谱条件下,盐酸小檗碱能够有效分离,平均回收率为98.3%,盐酸小檗碱在3.894~103.836μg/ml范围内与峰面积积分值线性关系良好,RSD=1.0%。结论改进后方法较原方法简便快捷,重复性、重现性好,结果准确。Objective To improve the determination method for determination ofberberine hydrochloride in Huanglian Shangqing tablets in volume I of Chinese Pharmacopoeia 2020.Methods The improved method was HPLC with Welch Welchrom C_(18)column(4.6 mm×250 mm,5μm)as the stationary phase,acetonitrile-0.033 mmol/L potassium dihydrogen phosphate(24∶76)as the mobile phase at a flow rate of 1.0 ml/min and a detection wavelength of 345.0 nm.Results Under the improved chromatographic conditions,berberine hydrochloride could be separated effectively,and the average recovery was 98.3%,and the linear range of berberine hydrochloride was 3.894~103.836μg/ml with the peak area integral value,and RSD was 1.0%.Conclusion The improved method is simpler,faster,more reproducible and accurate than the original method.
