清开灵颗粒减轻中暑小鼠肺部炎症反应及损伤

摘  要:目的:研究清开灵颗粒对小鼠中暑的治疗作用。方法:建立小鼠中暑模型,血常规检测药物治疗前后中暑小鼠淋巴细胞、中性粒细胞、红细胞积压、血红蛋白、血小板等指标变化。TUNEL染色和免疫组织化学检测各组小鼠肺组织凋亡情况。苏木精和伊红染色观察各组药物治疗后小鼠的肺泡病理学变化。结果:研究显示,清开灵药物处理后,实验小鼠淋巴细胞比率、中性粒细胞比率、血红蛋白等指标恢复至正常范围,肺组织炎症细胞浸润程度与凋亡率降低,中、高剂量的清开灵处理后肺组织凋亡因子半胱天冬酶的表达显著降低,且存在显著性差异。结论:清开灵颗粒能有效治疗中暑引起的肺部炎症和损伤,具备开发为抗中暑药物的前景。Objective:Study the therapeutic effect of Qingkailing granules on heatstroke in mice.Methods:Establish a mouse model of heat stroke,and detect changes in indicators such as lymphocytes,neutrophils,red blood cell accumulation,hemoglobin,and platelets in heat stroke mice before and after drug treatment using blood routine tests.TUNEL staining and immunohistochemistry were used to detect the apoptosis of lung tissue in each group of mice.Hematoxylin and eosin staining were used to observe the pathological changes in the alveoli of mice treated with various drugs.Results:Research has shown that after treatment with Qingkailing,the lymphocyte ratio,neutrophil ratio,hemoglobin and other indicators of experimental mice returned to normal range,and the degree of inflammatory cell infiltration and apoptosis rate in lung tissue decreased.The expression of apoptosis factor caspase in lung tissue was significantly reduced after treatment with medium and high doses of Qingkailing,and there was a significant difference.Conclusion:Qingkailing granules can effectively treat lung inflammation and injury caused by heatstroke.It has the prospect of being developed as anti-heatstroke drug.
