摘 要:目的 测定SA3、SA4和SA5三条短肽的抗菌活性并对其抗菌机制进行初步研究。方法 分析SA3、SA4和SA5三条短肽的理化性质,通过Fmoc固相合成方式制备三条短肽。通过最低抑菌浓度实验测定其抗菌活性。用N-苯基-1-萘胺和2-硝基苯基-β-D-吡喃半乳糖苷为探针进行三条短肽的细胞外膜、内膜渗透性实验。通过钙黄绿素泄露实验进一步验证三条短肽与细胞膜的作用。结果 SA3、SA4和SA5三条短肽对革兰阴性菌和革兰阳性菌均具有较强的杀伤作用。通过细菌外膜、内膜渗透性实验和钙黄绿素泄露实验证明细胞膜是抗菌肽的作用靶点。结论 SA3、SA4和SA5三条短肽通过作用于细胞膜产生抑制菌作用。Objective To measure the antibacterial activity of SA3,SA4,and SA5 short peptides and to conduct a preliminary study on their antimicrobial mechanisms.Methods The physicochemical properties of SA3,SA4,and SA5 short peptides were analyzed,and the peptides were synthesized using the Fmoc solid-phase method.Their antibacterial activity was assessed through the minimum inhibitory concentration(MIC)test.The permeability of the outer and inner bacterial membranes of the short peptides was investigated using N-phenyl-1-naphthylamine and 2-nitrophenyl-β-D-galactoside as probes.The interaction of the short peptides with the cell membrane was further validated by the calcein leakage assay.Results SA3,SA4,and SA5 short peptides exhibited strong bactericidal effects against both Gram-negative and Gram-positive bacteria.The cell membrane was confirmed as the target site of the antimicrobial peptides through outer and inner membrane permeability tests and the calcein leakage assay.Conclusions SA3,SA4,and SA5 short peptides exert their antibacterial effects by acting on the cell membrane.
