摘  要:为了保护纸质文物的历史价值,提出生物酶在纸质文物霉斑去除中的应用研究。所提方法为了明确待处理纸质文物表面的霉菌种类,对纸质文物样本实施擦拭采样后,利用快速微量提取法对菌株的DNA实施抽取,结合OTU聚类法明确霉菌种类,为后续清洗试剂中应用生物酶的选择奠定基础。在完成针对性除霉生物酶剂及匹配表活剂的种类选择后,利用虹吸清洗装置在霉斑模拟样品表面,实施清洗剂最佳配比的测试。通过不同AOS表活剂添加浓度下,样本纸张的拉力变化测试与色差变化测试,明确表活剂的最佳添加浓度,进一步优化生物酶复配清洗剂的除霉效果。In order to ensure the historical value of paper cultural relics,the application research of biological enzymes in the removal of mold spots on paper cultural relics is proposed.The proposed method aims to identify the types of fungi on the surface of ancient book paper pages to be processed.After wiping and sampling the paper cultural relics samples,the DNA of the strains is extracted using the rapid micro extraction method,and the OTU clustering method is used to identify the types of fungi,laying a foundation for the selection of biological enzymes in subsequent cleaning reagents.After completing the selection of targeted antifungal enzyme agents and matching surfactants,a siphon cleaning device was used to test the optimal ratio of cleaning agents on the surface of simulated samples with mold spots.Through testing the tensile strength change and color difference change of sample paper under different AOS surfactant addition concentrations,the optimal concentration of surfactants was determined to further optimize the antifungal effect of the enzyme complex cleaning agent.
