摘 要:试验旨在探究帕米尔高原绵羊低氧适应机制以及藏绵羊等其他动物是否有趋同性进化,为培育适应帕米尔高原低氧环境的塔什库尔干绵羊新品系提供分子辅助育种标记。试验以新疆平原(喀什)地区多浪羊为对照,采集塔什库尔干羊和马可波罗盘羊的样本,采用PCR扩增目的基因,琼脂糖凝胶电泳检测扩增产物并进行测序和单核苷酸多态性(SNP)位点分析。结果显示,46只多浪羊、18只塔什库尔干羊和7只盘羊的HIF-1α基因第10外显子有4个多态位点和5种单倍型,其中Hap2和Hap3为主要单倍型,分别位于盘羊和塔什库尔干羊的基因中。研究表明,塔什库尔干羊与盘羊基因多态位点的基因多态性与其对高原低氧环境的适应相关,其中G39210A位点突变等位基因(A)频率与海拔趋势呈正相关,故推测AA基因型与低氧适应有关。The aim of this experiment was to explore the mechanism of hypoxia adaptation in Pamir sheep and whether there is convergent evolution in other animals such as Tibetan sheep,in order to provide molecular markers for breeding new Tashkurgan sheep lines adapted to the low oxygen environment in Pamir Plateau.Using Duolang sheep in Xinjiang Plain(Kashgar)as the control,samples of Tashkurgan sheep and Marco Polo Argali were collected.PCR was used to amplify the target gene,and agarose gel electrophoresis was used to detect the amplification products,sequencing and single nucleotide polymorphism(SNP)site analysis.The results showed that there were four polymorphic loci and five haplotypes in exon 10 of HIF-1αgene in 46 Duolang sheep,18 Tashkurgan sheep and 7 Ovisammon,among which Hap2 and Hap3 were the main haplotypes,located in the genes of Ovisammonand Tashkurgan sheep,respectively.The results showed that the polymorphism of polymorphic loci of Tashkurgan sheep and Ovisammonwas related to their adaptation to low oxygen environment in the plateau,and the frequency of mutant allele(A)of G39210A was positively correlated with the altitude trend,so it was presumed that AA genotype was related to low oxygen adaptation.
