摘  要:目的 探讨搜风愈喘方及拆方对哮喘大鼠气道炎症及气道高反应性的作用机制。方法 将28只大鼠随机分为正常组、模型组、地塞米松组、搜风愈喘方组、祛伏风方组、消暗瘀方组、祛宿痰方组;除正常组外,其余组大鼠采用卵清白蛋白(ov-albumin, OVA)注射致敏及雾化激发复制哮喘大鼠模型,运用搜风愈喘方及拆方进行干预。使用酶联免疫法检测血清中白介素6(interleukin-6,IL-6)、白介素13(interleukin-13,IL-13)、转化生长因子β1(transforming growth factor β1,TGF-β1)、肿瘤坏死因子α(tumor necrosis factor alpha, TNF-α);苏木精—伊红染色及免疫组化法观察气管及肺组织病理形态;聚合酶联反应法检测大鼠肺组织匀浆中IL-6、IL-13、TGF-β1、TNF-α的mRNA表达;无创肺功能检测哮喘大鼠气道高反应性。结果 与正常组相比,模型组哮喘发作性反应明显,支气管及肺组织周围可见明显炎症细胞浸润,支气管管壁增厚;血清IL-6、IL-13、TGF-β1、TNF-α含量及mRNA表达显著升高(P<0.05)、气道反应性增强(P<0.05)。与模型组相比,地塞米松组、搜风愈喘方组、祛伏风方组、消暗瘀方组、祛宿痰方组大鼠哮喘表现减轻,气道及肺组织炎症细胞浸润减轻,支气管管壁轻度增厚;血清IL-6、IL-13、TGF-β1、TNF-α含量及mRNA表达可显著降低(P<0.05)、气道反应性显著降低(P<0.05)。各中药组组间比较,搜风愈喘方组在降低气道炎症、改善肺组织病理损伤、改善气道高反应性和减少血清IL-6、IL-13、TGF-β1、TNF-α含量及mRNA表达方面优于各拆方组(P<0.05)。结论 搜风愈喘方及拆方可降低IL-6、IL-13、TGF-β1、TNF-α表达,改善哮喘气道高反应性;祛伏风方组、消暗瘀方组、祛宿痰方组相比,祛伏风方组在改善气道炎症、降低气道高反应性方面具有优势。Objective To investigate the mechanism of Soufeng Yuchuan Decoction and dismantling decoction on airway inflammation and airway hyperresponsiveness in asthmatic rats.Methods Twenty-eight rats were randomly divided into the normal group,model group,dexamethasone group,Soufeng Yuchuan Decoction group,Qufufeng Decoction group,Xiaoanyu Decoction group and Qusutan Decoction group;in addition to the normal group,the rat model of asthma was established by sensitization with injection and atomization of ov-albumin(OVA),and Soufeng Yuchuan Decoction and dismantling decoction were used for intervention.The serum levels of interleukin-6(IL-6),interleukin-13(IL-13),transforming growth factorβ1(TGF-β1)and tumor necrosis factorα(TNF-α)were detected by enzyme-linked immunoassay(ELISA).HE staining and immunohistochemistry were used to observe the pathological morphology of airway and lung tissues.The mRNA expression of IL-6,IL-13,TGF-β1 and TNF-αin rat lung homogenate was detected by polymerase linked reaction(PCR).Airway hyperreactivity of asthmatic rats was detected by pulmonary function in vitro.Results Compared with the normal group,the asthmatic manifestations of rats in the model group were obvious,and a large number of inflammatory cells could be seen around the bronchus and lung tissues,the bronchial tube wall was thickened,the serum IL-6,IL-13,TGF-β1,TNF-αcontents and mRNA expression were significantly increased(P<0.05).Airway hyperreactivity was enhanced(P<0.05);compared with the model group,the asthma manifestations,inflammatory cell infiltration in airway and lung tissue in the dexamethasone group,Soufeng Yuhuan Decoction group,Qufufeng Decoction group,Xiaoanyu Decoction group and Qusutan Decoction group were decreased.The bronchial tube wall were slightly thickened.The levels of IL-6,IL-13,TGF-β1,TNF-αcontent and mRNA expression were significantly decreased(P<0.05).Airway hyperreactivity was significantly decreased(P<0.05);Compared with other Chinese medicine groups,Soufeng Yucuan Decoction group was super
