一规模化养殖场猪葡萄球菌的分离鉴定及其毒力、致病力研究

摘　　要：一规模化养殖场内10~40日龄仔猪发生渗出性皮炎,拟通过实验室诊断探明病因、确定病原。采集发病仔猪皮肤组织进行细菌分离,通过小鼠致死试验、仔猪毒力试验确定病原菌,对病原菌进行形态学观察、生化特性分析、毒力基因检测以及药敏分析。结果显示,经分离纯化获得1株分离株,其形态学与生化特性与猪葡萄球菌一致,命名为JS株。分离株以10~9 CFU/mL剂量经腹腔感染小鼠可全部致死,以10~8 CFU/mL剂量皮下感染仔猪能够复制出渗出性皮炎病例;葡萄球菌表皮脱落素Exh A与Shet A检测呈阳性,Exh B、Exh C、Exh D和Shet B检测阴性。药敏试验结果显示,分离株对头孢呋辛敏感,对头孢唑啉、头孢曲松和头孢氨苄中度敏感,对头孢哌酮、头孢拉定等14种抗生素呈现不同程度的耐药。研究表明,该养殖场引发仔猪渗出性皮炎的病原是猪葡萄球菌,分离株携带两种毒力基因并表现出多重耐药性。Exudative dermatitis occurred in piglets aged 10 to 40 days in a large-scale farm,and the cause and pathogen were determined by laboratory diagnosis.The skin tissue of infected piglets was collected for bacterial isolation,and the pathogenic bacteria were determined by mouse lethal test and piglet virulence test.The pathogenic bacteria were observed by morphology,analyzed by biochemical characteristics,detected by virulence gene and analyzed by drug susceptibility.The results showed that one isolated strain was obtained after isolation and purification,and its morphological and biochemical characteristics were consistent with that of Staphylococcus hyicus.It was named JS strain.All the mice infected by the isolated strain could be killed by peritoneal infection at the dose of 109 CFU/mL,and the piglets infected subcutaneously at the dose of 108 CFU/mL could reproduce exudative dermatitis cases.Staphylococcal epidermidin ExhA and ShetAwere positive,while ExhB,ExhC,ExhD,and ShetBwere negative.The results of drug sensitivity test showed that the isolated strain were sensitive to cefuroxime,moderately sensitive to cefazolin,ceftriaxone and cefalexin,and showed varying degrees of resistance to 14 kinds of antibiotics such as cefoperazone and cephaloadine.The study showed that the pathogen causing piglet exudative dermatitis in this farm was Staphylococcushyicus,and the isolates carried two virulence genes and showed multiple drug resistance.
