摘  要:为解决黑木耳栽培生产中存在的菌种老化退化问题,需研发出黑木耳菌种复壮的有效方法,以增强菌丝活力。本试验以已退化的黑木耳菌种为对象,研究PDA培养基中分别添加玉米粉、蛋白胨、螺旋藻粉、麦芽浸膏、牛肉膏、酵母粉共6种有机氮物质对退化黑木耳菌丝生长速度、生物量及其营养物质含量、活性氧含量、抗氧化酶活性等指标的影响。结果表明,相较于PDA培养基(CK),添加螺旋藻粉与麦芽浸膏培养基的菌丝生长速度快、生物量高,菌丝体蛋白质、多糖含量增加,且均达到显著水平;两处理菌丝体O_(2)-和H_(2)O_(2)含量较CK都大幅降低,除添加麦芽浸膏培养基的菌丝体H_(2)O_(2)含量降幅差异不显著外,其他均达显著水平;添加螺旋藻粉培养基的菌丝体过氧化物酶(POD)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性较CK都显著增加。综上所述,复壮黑木耳退化菌种的最佳培养基为添加螺旋藻粉培养基。本研究结果可为复壮黑木耳菌种老化退化菌种提供技术支持和理论参考。In order to solve the problem of stain degradation in cultivation and production process of black fungus(Auricularia auriculata),it is necessary to develop an effective method of fungus rejuvenation to enhance the viability of mycelia.In this experiment,the degraded black fungus strain was used as research ob-ject;the effects of adding six organic nitrogen substances,including corn flour,peptone,spirulina powder,malt extract,beef extract and yeast extract,on the mycelial growth rate,biomass,contents of nutrients and active oxygen,antioxidant enzymes activities of degraded black fungus mycelia in PDA medium were studied.The results showed that compared with PDA medium(CK),adding spirulina powder and malt extract made the mycelia grow faster,the biomass be higher,and the contents of protein and polysaccharide in mycelia in-crease significantly,but reduced the contents of O_(2)-and H_(2)O_(2)in mycelia significantly except the H_(2)O_(2)content in mycelia with the addition of malt extract.The activities of peroxidase(POD),superoxide dismutase(SOD)and catalase(CAT)in mycelia were significantly increased when adding spirulina powder compared with CK.To sum up,the optimal culture medium for rejuvenating the degraded strains of black fungus was PDA culture medium with spirulina powder added.This study could provide technical support and theoretical references for rejuvenating the aging and degraded black fungus strains.
