大白菜幼苗根系分泌物化学组分的鉴定

摘  要:为探究重茬培养条件下大白菜根系分泌物组分对植株生长的影响,为进一步阐明连作障碍机理提供理论依据,研究采用水培浸提大白菜幼苗根系分泌物,并借助气相色谱联用技术(GC-MS)分析和鉴定其根系分泌物的化学组分变化与差异;利用主成分分析(PCA)、偏最小二乘判别分析(PLS-DA)、代谢物热图分析和VIP图分析进行处理组间差异物质筛选。结果表明:在大白菜重茬培养液的根系分泌物中共鉴定出161种有机物,其中重茬液处理与营养液对照共有的成分是159种,重茬液处理和对照各有1个特异组分,分别为棕榈酸和4-乙酰苯甲酸。大白菜根系分泌物的PCA和PLS-DA分析结果表明,2组根系分泌物的物质成分差异显著,共筛选出47种差异化合物,种类较多的化合物有11种有机酸类、5种酰胺类、4种醇类、5种有机硅化物、3种糖类、3种胺类、2种烃类、2种氨基酸类以及2种维生素类。代谢物热图及VIP图分析表明,重茬液处理和对照根系分泌物相对含量均存在显著差异,推测重茬液处理根系分泌物中的棕榈酸、1-甲基苯乙烯、L-丙氨酸、半乳糖胺和1R,2S-苯丙胺是造成大白菜幼苗产生自毒作用的主要物质。To investigate the effects of root exudate components on plant growth of Chinese cabbage under continuous cropping conditions,and to provide theoretical basis for further elucidating the mechanism of continuous cropping obstacles,taking the root exudates of Chinese cabbage seedlings extracted from continuous cropping and single nutrient solution with hydroponics as materials,and the chemical composition changes and differences of their root exudates were analyzed and identified by using gas chromatography-mass spectrometry (GC-MS) technology.Principal component analysis (PCA),partial least squares discriminant analysis (PLS-DA),metabolite heatmap analysis and VIP plot analysis were used to screen out the difference substances between the two groups.The results showed that 161 types of organic compounds were identified in the root exudates of Chinese cabbage in continuous cropping liquid,of which 159 types of organic compounds were common to the continuous cropping liquid and the control group with nutrient solution.The continuous cropping liquid and control group each had one specific component,namely palmitic acid and 4-acetylbenzoic acid,respectively.The PCA and PLS-DA results of root exudates from Chinese cabbage showed significant differences in the material composition between the two groups.A total of 47 types of diverse compounds were screened,with 11 types of organic acids,5 types of amides,4 types of alcohols,5 types of organic silicides,3 types of sugars,3 types of amines,2 types of hydrocarbons,2 types of amino acids and 2 types of vitamins being more varieties of diverse compounds.Metabolite heatmap analysis and VIP map analysis showed that there were significant differences in the relative contents of root exudates between the continuous cropping liquid and the control group.It could be inferred that palmitic acid,1-methylstyrene,L-alanine,galactosamine and 1R,2S-phenylalanine in the root exudates of the continuous cropping liquid were the main substances causing autotoxicity in Chinese cabbage seedl
