摘  要:以最新的科研成果为基础设计了一个针对制药工程专业本科生的综合创新实验,研究了路易斯酸催化炔丙醇和2-烯基苯胺[4+2]环加成生成喹啉类化合物的反应,通过对催化剂、添加剂、溶剂、反应时间、反应温度等条件的考察,从而提高反应收率。在最优反应条件下考察带有不同取代基的炔丙醇和2-烯基苯胺的底物适用范围。该实验不仅可以提高学生的基本实验操作技能,加深对有机化学和药物合成的机理,同时能够培养学生的团队意识和创新精神。A comprehensive innovation experiment was designed for undergraduates majoring in Pharmaceutical Engineering on the basis of the latest scientific research results.The Lewis acid catalyzed[4+2]annulation reaction of propargylic alcohols with 2-vinylanilines for the assembly of quinoline skeletons was investigated.With the purpose of increasing the yield of this protocol,a series of reaction factors including catalyst,additive,solvent,reaction time and reaction temperature were investigated.The generalizability of the tandem annulation was explored by employing propargylic alcohols with 2-vinylanilines bearing various substituent groups under optimal reaction conditions.The undergraduate could not only improve their basic experimental operation skills,deepen their understanding of the organic chemistry and drug synthesis,but also cultivate their team consciousness,and innovative spirit through this comprehensive innovation experiment.
