摘 要:目的:以单因素实验为基础结合响应面法优化秦岭龙胆中龙胆苦苷(GGA)的提取工艺,并对其抗急性肝损伤活性进行研究。方法:以龙胆苦苷为评价指标,考察超声频率、提取时间、料液比对GGA提取率的影响,旨在确定最佳提取工艺。采用四氯化碳(CCl4)诱导小鼠急性肝损伤模型,检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)水平及肝组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量和肝脏重量。结果:GGA的最佳提取工艺条件为超声频率91 kHz、提取时间23 min、料液比1∶22,此工艺下的提取率达到0.328%,与预测值0.344%接近。与模型组比较,秦岭龙胆的高、中、低剂量组均可明显抑制小鼠血清中ALT、AST的升高(P<0.05),降低肝脏MDA含量,增强SOD活性(P<0.05)。结论:响应面法可用于GGA的提取工艺优化,此条件下秦岭龙胆提取物具有抗急性肝损伤的作用,本研究结果也可为秦岭龙胆的进一步开发利用提供理论依据。Objective:The extraction process of gentiopicroside from Gentiana apiata(GGA)was optimized by response surface method,and the anti-acute liver injury activity of G.apiata was determined.Methods:The factor levels were selected on the basis of single factor,and the response surface method was used to optimize the ultrasonic frequency,extraction time and solid-liquid ratio on the extraction of gentiopicroside from G.apiata.A mouse model of acute liver injury was induced by carbon tetrachloride(CCl4)to detect the levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum,as well as the superoxide dismutase(SOD)activity,malondialdehyde(MDA)content,and liver weight in liver tissue.Results:The optimal extraction conditions for GGA were ultrasonic frequency of 91 kHz,extraction time of 23 min,and solid-liquid ratio of 1∶22.Under these conditions,the extraction yield of gentiopicroside was 0.328%,which was close to the predicted value of 0.344%.Compared with the model group,the high-and low-dose groups of G.apiata could significantly inhibit the increase of ALT and AST in the serum of mice(P<0.05),reduce the liver MDA content,and enhance SOD activity(P<0.05).Conclusion:The response surface method can be used to optimize the extraction process of GGA.Under these conditions,the extract of G.apiata has the effect of anti-acute liver injury.The results of this study also provide a theoretical basis for the further development and utilization of G.apiata.
