摘 要:目的:建立同时测定蒲公英药材不同用药部位(全株、茎、叶)中羽扇豆醇、蒲公英甾醇、蒲公英萜醇3种成分含量的超高效液相色谱方法(UPLC),并比较差异。方法:采用WATERST3C18色谱柱(2.1mm×100mm,1.7μm),流动相:0.4%磷酸水溶液(A)-乙腈(B),3%A-97%B等度洗脱,柱温为25℃,流速为0.3 mL·min^(-1),进样量为10µL,检测波长为200 nm。结果:3种成分在各自线性范围内线性关系良好(r>0.999),精密度、重复性、稳定性(24 h)试验的RSD均小于2.0%(n=6),平均回收率大于97.11%,RSD<1.92%。10批样品结果显示不同产地蒲公英不同部位中3种萜类成分含量有所差异,安徽亳州产地全草中的羽扇豆醇和蒲公英甾醇含量最高,蒲公英萜醇在齐齐哈尔产地叶中含量最高。结论:该方法准确度高,重复性好,可为蒲公英的质量评价提供参考。Objective:An ultra performance liquid chromatography(UPLC)method was established for the simultaneous determination of lupeol,taraxosterol and taraxerol in different medicinal parts of Taraxacum mongolicum,and the discrepancies of 3 constituents were contrasted.Methods:The detection was conducted on a WATERS T3 C18 column(2.1 mm×100 mm,1.7μm),and 0.4%phosphoric acid aqueous solution(A)-acetonitrile(B)(3%A-97%B isocratic elution)was regarded as mobile phase for gradient elution.The temperature of the chromatographic column was maintained at 25℃.The flow rate was 0.3 mL·min^(-1),and the injection volume was 10µL.The detection wavelength was set at 200 nm.Results:The linear relationship between the three components was good in their linear ranges(r>0.999)and RSD value of precision,repeatability,and stability(24 h)tests were all less than 2.0%(n=6).The average recovery rate was outstrip than 97.11%,and RSD value was obtained by 1.92%.The results of 10 batches of samples showed that the contents of three terpenoids in different parts of T.mongolicum from different areas were different.The contents of lupeol and taraxasterol were the highest in the whole plant from Bozhou,Anhui,and the supreme content of taraxerol was found in the leaves of Qiqihar.Conclusion:The established UPLC method for the determination of the three components of T.mongolicum was accurate and reliable,which could provide reference for quality evaluation of T.mongolicum.
