摘 要:目的 探讨硫化氢(hydrogen sulfide,H_(2)S)对糖尿病(diabetes mellitus,DM)大鼠皮肤创面氧化应激的影响。方法 SPF级健康雄性SD大鼠18只,6周龄,体质量(210±10)g。随机选取12只,腹腔注射链脲佐菌素(streptozotocin,STZ)诱导糖尿病模型。随机血糖≥16.7μmol/L为建模成功。饲养3周后,将糖尿病大鼠随机分为模型组(DM)和NaHS处理组(NaHS)。其余6只SD大鼠为对照组(control)。手术切除各组大鼠背部皮肤,制备全层皮肤缺损模型。NaHS组大鼠每日腹腔注射NaHS溶液(56μmol/kg),DM组和control组大鼠每日腹腔注射等量0.9%生理盐水,连续注射21 d。术后第0、7、14、21天,检测皮肤创面愈合情况。术后21 d,取创缘组织检测氧化应激指标(ROS、MDA、SOD、CAT、T-AOC),TUNEL检测创面组织细胞凋亡情况。结果 与control组相比,DM组创面面积、创面组织中ROS、MDA含量以及细胞凋亡率显著升高(P<0.05),创面组织中SOD、CAT活性以及T-AOC水平显著降低(P<0.05)。与DM组相比,NaHS组创面面积、创面组织中ROS、MDA含量以及细胞凋亡率显著降低(P<0.05),创面组织中SOD、CAT活性以及T-AOC水平显著升高(P<0.05),但低于control组水平(P<0.05)。结论 H_(2)S可促进糖尿病大鼠皮肤创面愈合,其作用可能与减轻创面组织氧化应激,抑制细胞凋亡有关。Objective To explore the effect of hydrogen sulfide(H_2S) on oxidative stress in skin wounds of diabetic rats.Methods A total of 18 healthy 6-week-old male Sprague-Dawley rats(210±10 g) were selected.Twelve rats were intraperitoneally injected with streptozotocin(STZ) to induce a diabetic model.The random blood glucose levels≥16.7 mmol/L indicates successful modeling.After 3 weeks,diabetic rats were randomly assigned to either the model group(DM) or the NaHS treatment group(NaHS).The remaining 6 rats served as the control group(Control).Full-thickness skin defect models were created by surgically removing dorsal skin from all groups.Rats in the NaHS group received daily intraperitoneal injections of NaHS solution(56 μmol/kg),while the DM and Control groups received equivalent volumes of 0.9% saline,continuing for 21 days.The healing of skin wound was observed at 0 7,14 and 21 days after operation.On day 21 postoperatively,wound edge tissues were collected to measure oxidative stress indicators(ROS,MDA,SOD,CAT,T-AOC) and the cell apoptosis of the wound tissues were determined with the TUNEL kit.Results Compared to the Control group,the DM group exhibited significantly larger wound areas,increased ROS and MDA levels,and higher cell apoptosis rates(P<0.05),along with significantly reduced SOD and CAT activities and T-AOC levels(P<0.05).In contrast,the NaHS group showed significantly decreased wound areas,ROS and MDA levels,and cell apoptosis rates(P<0.05),as well as significantly increased SOD and CAT activities and T-AOC levels,although these did not reach the levels in the Control group(P<0.05).Conclusion H_(2)S can promote skin wound healing,which might be related to the reduction of oxidative stress and inhibition of apoptosis in diabetic rats.
