忧遁草醇提取物对四氯化碳诱导的小鼠急性肝损伤的保护作用

摘  要:目的 探究不同浓度的忧遁草醇提取物在对由四氯化碳导致的小鼠急性肝损伤中的保护作用及机制。方法 40只小鼠按照体重随机分为5组,其中包括正常组和模型组、给药组(10 mg/kg、20 mg/kg、40 mg/kg)。正常组和模型组均给予生理盐水连续灌胃,给药组给予不同浓度忧遁草醇提取物连续灌胃,共21 d。末次给药后,正常组腹腔注射橄榄油,其余组腹腔均注射含有0.1%四氯化碳橄榄油混合液建立小鼠急性肝损伤模型,16 h后小鼠眼眶采血,并分离血清。计算小鼠肝脏指数、测定丙二醛(Malondialdehyde,MDA)含量,检测血清中的谷氨酸氨基转移酶(Alanine aminotransferase,ALT)、天冬氨酸氨基转移酶(Aspartate aminotransferase,AST)、超氧化物歧化酶(Super-oxide dismutase,SOD)和谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)等指标。通过H&E染色和Masson染色检测小鼠肝脏的组织病理学变化。结果 给予忧遁草醇提取物后,小鼠肝指数及血清ALT、AST均显著低于四氯化碳造模后的小鼠(P<0.05);SOD、GSH-Px的含量均明显更高,而MDA的含量均降低(P<0.05);且呈剂量依赖性。在肝组织病理学检查中,不同浓度的忧遁草均不同程度缓解炎症细胞浸润和肝细胞肿胀、坏死。结论 忧遁草对急性肝损伤有一定的保护作用,其机制可能与抑制脂质过氧化反应和清除自由基有关。Objective To explore the protective effect of different concentrations of Clinacanthus nutans ethanol extract on carbon tetrachloride(CCl_4)-induced acute liver injury in mice and to elucidate its underlying mechanism.Methods Forty mice were randomly divided into five groups based on body weight,including control group,model group and three treatment groups receiving different concentrations of Clinacanthus nutans ethanol extract(10 mg/kg,20 mg/kg,40 mg/kg).The control group and the model group were given saline by continuous gavage,while treatment groups received the respective concentrations of ethanol extract daily for 21 days.Following the final administration,the control group was intraperitoneally injected with olive oil,and the remaining groups were injected with a 0.1% CCl_(4) mixture in olive oil to establish an acute liver injury model.16 hours later,blood was collected from the orbit of the mouse and the serum was separated.The liver index was calculated,the content of malondialdehyde(MDA),alanine aminotransferase(ALT),aspartate aminotransferase(AST),super-oxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in serum were measured.Histopathological changes of liver were detected by H&E and Masson staining.Results Administration of Clinacanthus nutans ethanol extract resulted in significantly lower liver indices and serum levels of ALT and AST compared to the CCl_4-induced injury group(P<0.05).The contents of SOD and GSH-Px were significantly higher,while the content of MDA was significantly lower(P<0.05).In pathological examinations,the treatment groups showed different degrees of relief of inflammatory cell infiltration and hepatocyte swelling and necrosis.Conclusion Clinacanthus nutans has a certain protective effect on acute liver injury,which may be attributed to its ability to suppress lipid peroxidation and eliminate free radicals.
