不同针刺手法对脑缺血再灌注大鼠眼针穴区肥大细胞活化作用的研究

摘　　要：目的探讨不同针刺手法对脑缺血再灌注(MCAO/R)大鼠眼针穴区肥大细胞活化作用的影响。方法35只大鼠随机分为空白对照组、模型对照组、无手法组、刮柄组、捻转组。采用改良线栓法创建MCAO/R模型,每12h干预1次,连续72h,干预前后采用改良Garcia评分法评分,处死后采用甲苯胺蓝对肥大细胞进行染色;ELISA法对穴区皮肤组胺(HA)、P物质(SP)、五羟色胺(5-HT)、前列腺素(PGD2)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)进行测定。采用SPSS26.0进行统计学分析。结果与模型对照组相比,无手法祖、刮柄组和捻转组神经功能评分降低,肥大细胞脱颗粒率提升,皮肤HA、SP、5-HT、PGD2表达提升,血清TNF-α、IL-6表达降低,差异有统计学意义(P<0.01);刮柄组和捻转组在HA、SP、5-HT、PGD2、TNF-α、IL-6表达方面有统计学意义(P<0.05)。结论刮柄法或捻转法对MCAO/R大鼠眼针穴区肥大细胞活化作用优于无手法。Objective To explore the effect of different acupuncture techniques on the activation of mast cells in the eye acupuncture point area of rats with cerebral ischemia-reperfusion(MCAO/R).Method Thirty-five rats were randomly divided into blank control group,model control group,no manipulation group,scraping handle group and twisting group.The MCAO/R model was created using an improved suture method,with intervention every 12 hours for 72 consecutive hours.The modified Garcia scoring method was used to score before and after intervention.Mast cells were stained with toluidine blue after euthanasia.The ELISA method was used to measure the levels of histamine(HA),substance P(SP),5-hydroxytryptamine(5-HT),prostaglandin D2,tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in the acupoint area skin.Statistical analysis was conducted using SPSS 26.0.Results Compared with those of the model control group,the nerve function score of the no manipulation group,scraping handle group and twisting group decreased.The degranulation rate of mast cells increased,the levels of skin HA,SP,5-HT and PGD2 increased,the levels of serum TNF-αand IL-6 decreased,and the difference was statistically significant(P<0.01).The expressions of HA,SP,5-HT,PGD2,TNF-αand IL-6 in scraping handle group and twisting group were statistically significant(P<0.05).Conclusion The scraping or twisting method has a better effect on the activation of mast cells in the eye acupuncture area of MCAO/R rats than no manual manipulation.
