摘 要:骨重建主要是由成骨细胞介导的骨形成与破骨细胞介导的骨吸收功能偶联并受到骨微环境如血管内皮细胞、免疫细胞等共同参与的动态过程,此过程是维持骨稳态的关键环节。在衰老或雌激素缺乏等病理条件下,骨重建失衡,骨微环境紊乱,导致骨代谢疾病如骨质疏松。骨重建相关细胞,如成骨细胞来源于骨髓间充质干细胞,破骨、血管内皮及免疫细胞来源于造血干细胞,这些细胞发育分化过程中形成不同亚型,异质性高,对调控骨重建发挥不同功能与作用。以肾主骨生髓中医理论,补肾中药是调控骨重建的主要药物,可调控骨重建多环节发挥作用。研究解析骨重建相关细胞的异质性,探讨补肾中药可能调控骨重建细胞异质性分化及功能作用,为探寻补肾中药治疗骨代谢疾病的机制与靶点以及阐释肾主骨生髓的新的细胞学机制提供方法与思路。Bone remodeling is a dynamic process that primarily involves the coupling of bone formation mediated by osteo-blasts and bone resorption mediated by osteoclasts,as well as the participation of bone microenvironments such as vascular endo-thelial cells and immune cells.This process is a key link in maintaining bone homeostasis.In pathological conditions such as ag-ing or estrogen deficiency,the dysregulation of bone remodeling disrupts the balance between bone formation and resorption,resul-ting in reduced bone mass and bone mineral density.This imbalance contributes to the development of bone metabolic disorders such as osteoporosis.Bone remodeling involves various cell types derived from bone marrow mesenchymal stem cells and hemato-poietic stem cells.These include osteogenic and osteoclastic cells,as well as vascular endothelial and immune cells.Throughout their development and differentiation,these cells give rise to different subtypes,exhibiting high heterogeneity and performing di-verse functions in the regulation of bone remodeling.According to the theory of kidney governing bone marrow in traditional Chi-nese medicine,kidney-tonifying Chinese medicine is considered the primary therapeutic approach to regulate bone remodeling.These medicines are believed to have the capacity to regulate various cells involved in bone remodeling.This study initiates with an exploration into the advancements in heterogeneity analysis of bone remodeling cells.It delves into the potential regulatory effects of kidney-tonifying Chinese medicine on the heterogeneous differentiation and functionality of bone remodeling cells.Furthermore,it investigates the mechanisms and targets underlying the treatment of bone metabolic disorders by kidney-tonifying Chinese medicine,offering methodologies and insights to elucidate the novel cellular mechanisms of kidney regulation over bone marrow.
