摘 要:目的分析断食对中波紫外线(UVB)照射诱导的光老化小鼠皮肤的影响。方法选取SPF级雄性C57BL/6小鼠18只,12周龄,体重18~22 g,采用随机数表法将其分为空白组、UVB组、UVB+断食组,每组6只。除空白组外,其余两组应用UVB照射制备小鼠皮肤光老化模型,以小鼠背部出现皮肤色素沉着加重,皮肤粗糙、增厚,褶皱明显等体征,提示造模成功。UVB+断食组同时进行断食。12周后处死小鼠,HE染色和EVG-Weigert染色观察皮肤形态学改变,检测皮肤和血清中超氧化物歧化酶(SOD)、丙二醇(MDA)、羟脯氨酸(HYP)含量变化。结果与空白组比较,UVB组小鼠背部皮肤出现明显的色素沉着、粗糙、增厚等光老化改变;与UVB组比较,UVB+断食组小鼠背部光老化改变减轻。除空白组外,两组均表现出明显的局部炎症浸润现象,而UVB+断食组的浸润现象相对较轻。与空白组比较,UVB组胶原纤维断裂、紊乱,部分弹力纤维增生、变粗、缠结;与UVB组比较,UVB+断食组小鼠胶原纤维排列整齐。与空白组比较,UVB组小鼠表皮和真皮厚度增加(P<0.05);与UVB组比较,UVB+断食组小鼠真皮和表皮厚度变薄(P<0.05)。与空白组比较,UVB组小鼠弹力纤维面积增加(P<0.05);与UVB组比较,UVB+断食组小鼠弹力纤维面积减小(P<0.05)。与空白组比较,UVB组小鼠皮肤SOD活性降低、MDA含量升高、HYP含量下降(P<0.05);与UVB组比较,UVB+断食组小鼠皮肤SOD活性升高、MDA含量降低(P<0.05);UVB组与UVB+断食组HYP含量比较,差异无统计学意义(P>0.05)。与空白组比较,UVB组小鼠血清SOD活性下降、MDA含量升高、HYP含量下降(P<0.05);与UVB组比较,UVB+断食组小鼠血清SOD活性升高、MDA含量降低、HYP含量升高(P<0.05)。结论断食可提高皮肤组织抗氧化能力,有效改善皮肤因紫外线照射诱导的表皮、真皮增厚,真皮结缔组织排列紊乱,纤维断裂等问题,对皮肤老化具有一定保护作用,可为临床上皮肤老�Objective To analyze the effect of fasting on the skin of Ultraviolet B(UVB)-induced photoaging mice.Methods Eighteen SPF grade male C57BL/6 mice,aged 12 weeks and weighing 18-22 g were selected.They were divided into blank group,UVB group,and UVB+fasting group by random number table method,with six mice in each group.Except for blank group,the mice skin photoaging model was prepared by UVB irradiation in the other two groups.The signs such as increased skin pigmentation,rough skin thickening,and obvious wrinkles on the back of the mice indicated that the modeling was successful.The UVB+fasting group underwent fasting at the same time.After 12 weeks,the mice were killed,and the skin morphological changes were observed by HE staining and EVG-Weigert staining,the contents of superoxide dismutase(SOD),malondialdehyde(MDA),and hydroxyproline(HYP)in skin and serum were detected.Results Compared with blank group,the UVB group showed obvious pigmentation,roughness,and thickening,and other photoaging changes;compared with UVB group,UVB+fasting group reduced the change of dorsal photoaging.Except the blank group,both groups showed obvious local inflammatory infiltration,and the UVB+fasting group showed relatively mild infiltration.Compared with blank group,collagen fibers in UVB group were broken and disorganized,and some elastic fibers were hyperplasia,thickening,and entangling;compared with UVB group,collagen fibers in UVB+fasting group were arranged neatly.Compared with blank group,the epidermal and dermal thickness of UVB group was increased(P<0.05);compared with UVB group,the thickness of dermis and epidermis of UVB+fasting group was thinner(P<0.05).Compared with blank group,the area of elastic fibers of UVB group was increased(P<0.05);compared with UVB group,the area of elastic fibers of UVB+fasting group was reduced(P<0.05).Compared with blank group,SOD activity was decreased,MDA content was increased,and HYP content was decreased in skin of UVB group(P<0.05);compared with UVB group,SOD activity was increased and M
