摘  要:目的探究神经调节素-1β(NRG-1β)对心力衰竭(HF)大鼠心脏交感神经系统的影响。方法选取31只成年Wistar雄性大鼠,将其分为对照组(7只)和模型组(24只)。对照组皮下注射0.5 ml生理盐水,模型组皮下注射340 mg/kg异丙肾上腺素,间隔24 h,重复注射1次。8周后模型组存活的15只大鼠分为HF组(7只),NRG-1β组(8只)。NRG-1β组尾静脉注射NRG-1β100μg/kg,2次/周,连续4周;对照组、HF组尾静脉注射生理盐水。比较各组左室收缩末期内径(LVDs)、左室舒张末期内径(LVDd),左室射血分数(LVEF)、左室短轴缩短率(LVFS);比较各组HE染色、酪氨酸羟化酶(TH)免疫组织化学染色结果;比较各组心脏去甲肾上腺素(NE)含量及β-肾上腺素能受体(AR)亚型m RNA表达。结果与对照组比较,HF组LVDs、LVDd扩大,LVFS、LVEF降低(P<0.05)。与HF组比较,NRG-1β组LVDs、LVDd缩小,LVFS、LVEF升高(P<0.05)。对照组心肌纤维排列整齐,细胞核正常;HF组心肌出现局灶性凝固性坏死;NRG-1β组心肌坏死较HF组减少。对照组心脏可见TH染色;与对照组比较,HF组心脏TH染色减少;与HF组比较,NRG-1β组心脏TH染色增加。与对照组比较,HF组心脏NE含量降低(P<0.05);与HF组比较,NRG-1β组心脏NE含量增加(P<0.05)。与对照组比较,HF组心脏β1-AR m RNA表达减少,β3-AR m RNA表达增加(P<0.05)。与HF组比较,NRG-1β组心脏β1-AR m RNA表达增加,β3-AR m RNA表达降低(P<0.05)。HF组与对照组心脏β2-AR m RNA表达比较,差异无统计学意义(P>0.05);NRG-1β组与HF组心脏β2-AR m RNA表达比较,差异无统计学意义(P>0.05)。结论NRG-1β纠正HF大鼠心脏交感神经系统异常,改善心脏结构与功能。Objective To investigate the effect of neuregulin-1β(NRG-1β)on cardiac sympathetic nervous system in rats with heart failure(HF).Methods A total of 31 adult Wistar male rats were selected,and they were divided into control group(7 rats)and model group(24 rats).The control group was injected with 0.5 ml of normal saline subcutaneously,and the model group was injected with 340 mg/kg isoprenaline subcutaneously,the injection repeat at an interval of 24 h.The 15 surviving rats after 8 weeks in the model group were divided into HF group(7 rats)and NRG-1βgroup(8 rats).NRG-1βgroup was given NRG-1β100μg/kg intravenously,twice a week for four consecutive weeks;control group and HF group were injected with normal saline through tail vein.Left ventricular systolic diameter(LVDs),left ventricular diastolic diameter(LVDd),left ventricular ejection fractor(LVEF),and left ventricular fractional shortening(LVFS)among all groups were compared;HE staining and immunohistochemical staining for tyrosine hydroxylase(TH)among all groups were compared;and the cardiac norepinephrine(NE)content andβ-adrenergic receptor(AR)subtype mRNA expression were compared among all groups.Results Compared with the control group,LVDs and LVDd were enlarged,and LVFS and LVEF were decreased in HF group(P<0.05);compared with HF group,LVDs and LVDd were decreased,and LVFS and LVEF were increased in NRG-1βgroup(P<0.05).In control group,the myocardial fibers were arranged neatly and the nucleus was normal;focal coagulation necrosis was showed in the myocardium of HF group;and myocardial necrosis in NRG-1βgroup was less than that in HF group.TH staining was observed in the heart of control group;compared with control group,the cardiac TH staining in HF group was decreased;and cardiac TH staining was increased in NRG-1βgroup compared with HF group.Compared with control group,the cardiac NE content in HF group was decreased(P<0.05);compared with HF group,the cardiac NE content in NRG-1βgroup was increased(P<0.05).Compared with control group,the cardi
