摘 要:为研究成纤维细胞生长因子2(FGF2)对鸡原代成肌细胞增殖和分化的影响,试验以鸡原代成肌细胞为研究对象,体外培养使其增殖并分化为肌管。在成肌细胞中添加不同浓度的FGF2重组蛋白,通过EdU和CCK8技术检测FGF2对原代成肌细胞增殖的影响,通过荧光定量PCR检测分化因子MYOD、MYOG的表达情况,分析FGF2对成肌细胞分化的影响。进一步通过转录组测序分析FGF2处理组和对照组的基因表达差异,挖掘调节肌肉发育的潜在分子机制。结果显示:添加FGF2重组蛋白能够显著促进成肌细胞增殖,抑制成肌细胞分化,在5 ng/mL时对细胞增殖促进效果最强。转录组测序数据显示,在FGF2处理组和对照组中存在424个差异表达基因,GO富集分析结果显示差异基因富集到细胞增殖、分化等通路,KEGG分析结果显示差异基因富集到MAPK通路。研究表明,FGF2可能通过影响MAPK通路进而调控成肌细胞的增殖和分化。To investigate the effect of FGF2 on the proliferation and differentiation of chicken primary myoblasts,the experiment focused on chicken primary myoblasts,which were cultured in vitro to proliferate and differentiate into myotubes.FGF2 recombinant protein with different concentrations were added to myoblasts,and the effects of FGF2 on the proliferation of primary myoblasts were detected by using EDU and CCK8 techniques.Real-time quantitative PCR was used to detect the expression of differentiation factors MYOD and MYOG.In further,RNA-seq was used to analyze the gene expression differences between the FGF2 treatment group and the control group to explore potential molecular mechanisms regulating muscle development.The results showed that FGF2 recombinant protein could significantly promote myoblast proliferation,inhibit myoblast differentiation,and had the strongest effect on cell proliferation at 5 ng/mL.Transcriptome sequencing data showed that there were 424 differentially expressed genes between the FGF2 treatment group and control group.GO enrichment analysis showed that differentially expressed genes were enriched in pathways such as cell proliferation and differentiation,and KEGG analysis showed that differentially expressed genes were enriched in the MAPK pathway.The results indicated that FGF2 might regulate the proliferation and differentiation of myoblasts by affecting the MAPK pathway.
