摘 要:为探究PRKCD在鸡脂肪沉积和肌肉发育中的作用,研究以S3系、F系和H系3个品系0、2、5、10和15周龄鸡的肝脏、胸肌、腿肌、腹脂和皮脂组织以及增殖期和分化期的脂肪细胞和成肌细胞为试验材料,荧光定量PCR(Q-PCR)检测PRKCD的表达变化。结果显示:PRKCD在不同品系鸡的肝脏、胸肌、腿肌、腹脂、皮脂组织及细胞中均有表达,呈现出明显的品系和组织差异性。肝脏组织中,0、2、10和15周龄时,H系的表达量是最高的,显著高于S3系和F系(P<0.05);在胸肌和腿肌上,在2、5、10和15周龄时,S3系的表达量显著高于F系和H系(P<0.05);在皮脂上,2、5和15周龄时,F系的表达量最高,2和15周龄显著高于其他品系(P<0.05),5周龄时显著高于S3系(P<0.05);在腹脂上,5和15周龄时,F系的表达量显著高于其他品系(P<0.05);在成肌细胞和肌内脂肪细胞中,PRKCD增殖期50%表达显著高于分化期(P<0.05);PRKCD表达与S3系鸡的腿肌率显著正相关(P<0.05)。研究表明,PRKCD的表达呈现显著的品系和组织差异性,并与腿肌率呈显著正相关,该基因参与了肌肉发育和肌内脂肪沉积的调控过程,可作为鸡肌肉发育和肌内脂肪沉积新的候选基因。In order to investigate the role of PRKCD in chicken fat deposition and muscle development,the study utilized liver,pectoral muscle,leg muscle,abdominal fat and sebaceous tissues from three strains of chickens:S3,F,and H.Additionally,adipocytes and adult muscle cells at the proliferative and differentiation stages were included.Samples were collected at 0,2,5,10,and 15 weeks of age for each strain,and the changes in PRKCD expression were detected by fluorescence quantitative PCR(Q-PCR).The results showed that:PRKCD was expressed in liver,breast muscle,leg muscle,abdominal fat and sebaceous tissues and cells of different strains of chickens,showing obvious strain and tissue variability.In liver,the expression of H line was the highest and significantly higher than that of S3 line and F line at the age of 0,2,10 and 15 weeks(P<0.05).In breast muscle and leg muscle,the expression of PRKCD in S3 line was the highest and significantly higher than that in S3 line and F line at the age of 2,5,10 and 15 weeks.The expression of PRKCD in S3 line was the highest at the age of 2,5,10 and 15 weeks,and the expression of PRKCD in S3 line was the highest at the age of 2,5,10 and 15 weeks(P<0.05);In sebaceous muscle,at 2,5 and 15 weeks of age,the expression of F line was the highest,significantly higher than that of the other lines at 2 and 15 weeks of age(P<0.05),and significantly higher than that of S3 line at 5 weeks of age(P<0.05);In ventral sebaceous muscle,at 5 and 15 weeks of age,the expression of PRKCD in F line was significantly higher than that of the other lines(P<0.05);In pectoral muscle,at 5 and 15 weeks of age,the expression of PRKCD in F line was significantly higher than that of the other lines(P<0.05);In adult myoblasts and intramuscular adipocytes,the 50%expression of PRKCD was significantly higher in the proliferative stage than that in the differ‐entiation stage(P<0.05);PRKCD expression was significantly positively correlated with the rate of hamstrings in S3 line chickens(P<0.05).In conclusion,the expressio
