摘 要:本研究旨在开发一种高效分析化妆品中四种常见脂肪酸含量的方法,为此,建立了一种基于液相色谱-串联质谱技术的分析方法,该方法具有简便快捷的特点。具体操作如下:使用安捷伦ZORBAX Eclipse Plus C18RRHD液相色谱柱(2.1*50 mm,1.8μm),在30℃的柱温条件下进行分析,流动相由甲醇(A相)和0.005 mol/L乙酸铵水溶液(B相)组成,采用85%的A相和15%的B相进行等度洗脱,采用电喷雾电离(ESI)负离子模式。数据采集采用选择离子监测(SIM)模式,监测离子分别为月桂酸的199 m/z、肉豆蔻酸的277.1m/z、棕榈酸的255.2 m/z和油酸的283.3 m/z,通过外标法进行定量分析,确保了结果的准确性。结果表明,四种脂肪酸在10 min内能够有效分离,且三组相邻脂肪酸的分离度均大于1.5。检测限和定量限分别低至0.01μg/mL和0.05μg/mL,线性关系良好,相关系数R~2均达到0.999以上,在准确度方面,基质空白加标回收率介于91.2%至98.3%之间,重复性和精密度均表现良好。综上所述,本方法具有检出限低、精密度高的优点,能够快速且准确地检测化妆品中四种常见脂肪酸的含量,为行业的检测方法提供有效的技术参考。This study aims to develop an efficient method for analyzing the content of four common fatty acids in cosmetics.To this end,a liquid chromatography tandem mass spectrometry based analysis method was established,which has the characteristics of simplicity and speed.The specific operation is as follows:Use an Agilent ZORBAX Eclipse Plus C18 RRHD liquid chromatography column(2.1*50mm,1.8μm)for analysis at a column temperature of 30℃.The mobile phase consists of methanol(phase A)and 0.005 mol/L ammonium acetate aqueous solution(phase B).Equivalently elute with 85%phase A and 15%phase B,the negative ion mode of electric spray ionization(ESI)is adopted.The data was collected using the Selective Ion Monitoring(SIM)mode,with monitoring ions of 199 m/z for lauric acid,277.1 m/z for myristic acid,255.2 m/z for palmitic acid,and 283.3 m/z for oleic acid.Quantitative analysis was performed using an external standard method to ensure the accuracy of the results.The results showed that the four types of fatty acids could be effectively separated within 10 minutes,and the separation degrees of the three adjacent fatty acids were all greater than 1.5.The detection limit and quantification limit are as low as 0.01μg/mL and 0.05μg/mL,respectively.The linear relationship 2 is good,and the correlation coefficient R is above 0.999.In terms of accuracy,the matrix blank spiked recovery rate ranges from 91.2%to 98.3%,and the repeatability and precision are both good.In summary,this method has the advantages of low detection limit and high precision,and can quickly and accurately detect the content of four common fatty acids in cosmetics,providing effective technical reference for industry detection methods.
