摘 要:为研究灵芝孢子油的抗衰老功效,开展了生化实验、体外细胞实验和人体临床实验。生化实验测定灵芝孢子油对弹性蛋白酶的抑制率。细胞实验以人表皮细胞为研究对象,建立体外损伤模型,通过灵芝孢子油抑制细胞活性氧的能力来评价其抵抗UVB损伤紧致及抗皱的效果。人体临床实验测试志愿者使用样品前后皮肤水分含量(Moisture Measurement Value,MMV)、经皮失水率(Trans Epidermal Water Loss,TEWL)、弹性值(R2)、皮肤表面红斑指数(Erythema index,EI),并用VISIA-CR面部图像分析系统进行面部图像采集。试验结果显示,生化实验中灵芝孢子油在质量浓度为1%、0.5%、0.1%对弹性蛋白酶有显著的抑制作用。细胞实验中灵芝孢子油在质量浓度为0.01%、0.05%时,具有抑制ROS的作用,有一定的紧致抗皱效果。人体临床实验表明在凝露基质中添加质量分数为0.1%的灵芝孢子油,连续使用28天,志愿者MMV值增加了14.15%、TEWL值下降了17.57%、R2值增加了16.31%、EI值下降了13.01%。To study the anti-aging effect of Ganoderma spore oil,biochemical experiments,in vitro cell experiments,and human clinical experiments were conducted.Biochemical experiments were conducted to determine the inhibitory rate of Ganoderma spore oil on elastase.Cell experiments were conducted on human epidermal cells to establish an in vitro injury model.The ability of Ganoderma spore oil to inhibit cell reactive oxygen species was evaluated to evaluate its resistance to UVB induced tightening and wrinkle resistance.In human clinical trials,volunteers were tested for skin moisture measurement value(MMV),transdermal water loss(TEWL),elasticity value(R2),and skin surface erythema index(EI) before and after using the sample.Facial images were collected using the VISIA-CR facial image analysis system.The experimental results showed that Ganoderma spore oil had a significant inhibitory effect on elastase in biochemical experiments at mass concentrations of 1 %,0.5 %,and 0.1 %.In cell experiments,Ganoderma spore oil has an inhibitory effect on ROS and a certain tightening and anti wrinkle effect at mass concentrations of 0.01 % and 0.05 %.Human clinical experiments have shown that adding 0.1 % Ganoderma spore oil to the gel matrix and using it continuously for 28 days,the MMV value of volunteers increased by 14.15 %,TEWL value decreased by 17.57 %,R2 value increased by 16.31 %,and EI value decreased by 13.01 %.
