河南部分地区奶牛肠道纤毛虫感染情况及分子鉴定

摘  要:为了明确奶牛肠道纤毛虫的感染情况及其人兽共患风险,从河南省不同地区8个集约化奶牛场中采集462份新鲜粪样,采用粪便直接涂片法镜检和基于18S rDNA和ITS1-5.8S rDNA-ITS2序列对样品中纤毛虫感染情况和分子特性进行了鉴定。结果显示,奶牛粪便中存在有槽巴克斯顿纤毛虫和结肠小袋纤毛虫,其中有槽巴克斯顿纤毛虫感染率为29.9%,成年牛感染率显著高于其他年龄段奶牛(P<0.01);结肠小袋纤毛虫感染率为3.9%,不同年龄段感染率差异不显著(P>0.05),8份样品为有槽巴克斯顿纤毛虫和结肠小袋纤毛虫混合感染。未从奶牛粪便中检出瘤胃内纤毛虫,说明奶牛瘤胃内的共生纤毛虫不随粪便排出体外。研究结果为集约化奶牛场纤毛虫病的防控提供了重要的基础数据。In order to determine the infection of intestinal ciliates and the risk of zoonosis in dairy cows,462 fresh fecal samples were collected from 8 intensive dairy farms in different areas of Henan province.The infection and molecular characteristics of ciliates in the samples were identified by direct fecal smear microscopic examination and based on 18S rDNA and ITS1-5.8S rDNA-ITS2 sequences.The results showed that there were Buxtonella sulcata and Balantidioes coli in cow feces,in which the positive rate of Bu.sulcata was 29.9%.The positive rate of adult cattle was significantly higher than that of other ages(P<0.01);The positive rate of Ba.coli was 3.9%,and there was no significant difference among different ages(P>0.05),Eight samples were mixed infection of Bu.sulcata and Ba.coli.The rumen ciliates were not detected in the feces of dairy cows,indicating that the symbiotic ciliates in the rumen of dairy cows were not excreted with the feces.The results provide important basic data for the prevention and control of ciliosis in intensive dairy farms.
