摘 要:动物流感病毒具有跨种属传播引起人类感染甚至导致大流行的潜力,对公共卫生和社会发展带来挑战和威胁。流感病毒血凝素(hemagglutinin,HA)蛋白与宿主细胞唾液酸(sialic acid,SA)受体的结合是病毒入侵宿主的第一步,也是病毒跨种属传播的第一道屏障。流感病毒HA受体结合区域氨基酸的突变,会导致病毒受体结合特异性发生改变,是造成病毒跨种属传播的分子基础。本文总结了流感病毒SA受体的发现和常见检测方法、主要种类和结构、分布以及HA蛋白识别宿主受体的分子特征等关键决定因素,为阐明流感病毒的跨种属传播机制提供科学依据。Animal influenza A viruses are considered to have the potential to spread cross-species barrier,posing challenges and threats to public health and social development.The binding of influenza virus hemagglutinin(HA) protein to sialic acid(SA) receptors on host cells is the first step to entry and infection host cells,which considered to be the first step to cross-species transmission.Specific amino acid mutations at the viral HA receptor binding site lead to a shift in receptor-binding preference,which is the molecular basis and plays a key role for the occurrence of jumping the species barrier of influenza A viruses(IAVs).In this review,we summarize the discovery of IAVs SA receptors,the methods to assess receptor-binding specificity,the glycan types and structures of SA receptor,SA receptor distribution and the molecular basis of viral HA binding to host receptors,in order to provide a scientific theoretical basis for elucidating the cross-species barrier of IAVs.
