摘 要:为明确大连地区甜樱桃叶斑病的病原菌种类,采集病叶进行组织分离,观察病菌形态特征,结合rDNA-ITS、EF-1α、Alt a1基因序列分析对其种类进行鉴定,并经柯赫氏法则证实其致病性,发现引起大连地区甜樱桃叶斑病的病原菌为链格孢菌(Alternaria alternata)和细极链格孢菌(Alternaria tenuissima),分离频率分别为87.28%和12.72%。测定不同药剂对优势病原菌链格孢菌的室内毒力,供试的化学杀菌剂以戊唑醇抑菌效果最好,EC_(50)为0.3687 mg/L,氟菌·肟菌酯抑菌效果次之,EC_(50)为0.8582 mg/L;供试的生物杀菌剂以多抗霉素抑菌效果最好,EC_(50)为3.1494 mg/L,中生菌素抑菌效果次之,EC_(50)为3.3365 mg/L。研究结果可为大连地区甜樱桃叶斑病的诊断与防治提供理论依据。In order to clarify the pathogen of sweet cherry leaf spot in Dalian,we collected the diseased leaves for tissue separation and observed the morphological characteristics of the pathogen.The species were identified through rDNA-ITS,EF-1α,and Alt a 1 gene sequence analysis and their pathogenicity was confirmed through the Koch′s postulates.The results showed that the pathogens responsible for sweet cherry leaf spot in Dalian were Alternaria alternata and Alternaria tenuissima,with isolation frequencies of 87.54%and 12.46%,respectively.The toxicity test demonstrated that tebuconazole and fluopyram+trifloxystrobin had the strongest toxicity against Alternaria alternata,with EC_(50) values of 0.3687 mg/L and 0.8582 mg/L,respectively.Among the tested biocides,Polyoxin and Zhongshengmycin were the most effective biocides against A.alternata with EC_(50) values of 3.1494 mg/L and 3.3365 mg/L,respectively.The results of this study provide a theoretical basis for the diagnosis and integrated control of sweet cherry leaf spot in Dalian.
