摘 要:目的探讨肺维康颗粒对肺纤维化(PF)大鼠的影响及作用机制。方法将36只SD雄性大鼠随机分为空白组、模型组、吡非尼酮组和肺维康颗粒高、中、低剂量组,每组6只。除空白组外,余均用博来霉素气管内给药法造模。造模成功后,根据组别给予对应药物灌胃28 d,处死、取样。观察各组肺组织病理学改变,白细胞介素(IL)-6、转化生长因子(TGF)-β1含量和TGF-β1、SMAD3、Fibronectin蛋白表达。结果相较空白组,模型组染色可见更多结构损伤与胶原沉积,而吡非尼酮组与肺维康颗粒各组情况较模型组减轻。模型组IL-6、TGF-β1含量及TGF-β1、SMAD3、Fibronectin蛋白表达较空白组明显上调,吡非尼酮组与肺维康颗粒各组的上述指标则均较模型组降低,差异均有统计学意义(P<0.01或P<0.05)。结论肺维康颗粒可减少PF大鼠炎症因子释放、延缓疾病进展,其作用机制可能与调控TGF-β1/SMAD3信号通路相关。Objective To investigate the effects and mechanisms of Feiweikang granules on pulmonary fibrosis rats.Methods Thirtysix male SD rats were randomly divided into blank group,model group,pirfenidone group,and high,medium,low dose groups of Feiweikang granules,with six rats in each group.Except for the blank group,bleomycin was intratracheally administered to modeling.After successful modeling,corresponding drugs were administered according to the groups,and the rats were euthanized after continuous gavage for 28 days.The histopathological changes in lung tissues were observed,and the IL-6 and TGF-β1 content and TGF-β1,SMAD3,fibronectin protein expressions were determined.Results Compared with the blank group,the model group showed more structural damage and collagen deposition visible in staining.The conditions were alleviated in the pirfenidone group and the Feiningkang granules groups compared to the model group.The IL-6 and TGF-β1 levels in the model group,as well as the TGF-β1,SMAD3,fibronectin protein expressions were significantly upregulated compared to the blank group(P<0.01).The IL-6 and TGF-β1 levels,along with the TGF-β1,SMAD3,fibronectin protein expressions were decreased in the pirfenidone group and Feiweikang granules groups compared to the model group(P<0.01 or P<0.05).Conclusion Feiweikang granules can reduce the release of inflammatory factors,delay the progression of pulmonary fibrosis,its mechanism may be related to the regulation of TGF-β1/SMAD3 signaling pathway.
