摘  要:蔗糖合成酶(SUS)是植物蔗糖代谢中极其重要的一种酶,既可以催化蔗糖的分解,又可以催化其合成。SUS由多基因家族编码,目前在多个物种中已分离出SUS基因家族全部成员,但香蕉中至今还没有SUS基因家族的报道。本研究利用香蕉基因组,鉴定出5个SUS基因,分别命名为MaSUS1~5,不均匀地分布于Chr3、Chr7、Chr8、Chr9等4条染色体上,其氨基酸数量为624~844aa,蛋白二级结构均主要由α-螺旋和无规则卷曲组成。亚细胞定位预测分析表明,5个SUS基因均位于细胞质中;进化树分析发现,MaSUS1和MaSUS4属于SUSⅡ亚家族,MaSUS2和MaSUS5属于SUSⅢ亚家族,MaSUS3属于SUSⅠ亚家族;对其基因结构进行分析发现,外显子数量为12~15,内含子数量为11~14;对其顺式作用元件进行预测发现,光响应元件数量和种类最多,其次是激素响应元件,逆境响应元件最少。本研究可为研究调控香蕉蔗糖代谢的分子机制提供前期基础。Sucrose synthase(SUS)is a crucial enzyme in plant sucrose metabolism that can catalyze both the decomposition and synthesis of sucrose.SUS is encoded by multiple gene families,and all members of the SUS gene family have been iso-lated from several species.However,there have been no reports on the SUS gene family in bananas.Five SUS genes,named MaSUS1-5,were identified from the banana genome in this study and were unevenly distributed on four chromosomes,in-cluding Chr3,Chr7,Chr8,and Chr9.Its amino acid numbers range from 624 aa to 844 aa.Random coils andα-helixes were the main secondary structures.All five SUS genes were located in the cytoplasm by subcellular localization prediction analysis.Evolutionary tree analysis revealed that MaSUS1 and MaSUS4 belong to the SUS II subfamily,MaSUS2 and MaSUS5 belong to the SUS III subfamily,and MaSUS3 belongs to the SUS I subfamily.The number of exons ranged from 12 to 15,and the number of introns ranged from 11 to 14.The prediction of cis-acting elements revealed that the number of light-responsive elements was the highest,followed by hormone-responsive elements,and that of stress-responsive elements was the lowest.This study provides a preliminary basis for studying the molecular mechanisms of regulating sucrose metabolism in bananas.
