摘 要:以青海省门源县采集的中国沙棘(Hippophae rhamnoides subsp.sinensis Rousi)成熟叶片为试材,采用单因素试验和响应面分析法,研究了中国沙棘叶黄酮的提取和纯化工艺,优化其提取效率和纯化效果,以期为中国沙棘叶片黄酮类化合物的有效利用提供参考依据。结果表明:在75%乙醇、81℃温度、110 min提取时间以及料液比1∶8 mg·mL^(-1)的条件下进行2次醇提取,其理论黄酮得率为60.56 mg·g^(-1),实测结果为(60.52±0.18)mg·g^(-1);以D101大孔树脂作为吸附剂,系统的优化筛选上样量、浓度、酸碱环境、洗脱剂用量、洗脱速率及洗脱剂浓度等参数,得到D101大孔树脂吸附率为91.19%±0.32%,解析率为92.27%±0.75%,使黄酮含量提高了近2.95倍,且吸附率高于以往的报道。The mature leaves of Hippophae rhamnoides subsp.sinensis Rousi collected from Menyuan county,Qinghai Province,were used as the experimental material.A single-factor experiment and response surface methodology were employed to investigate and optimize the extraction and purification process of flavonoids from Hippophae rhamnoides leaves and to enhance the extraction efficiency and purification outcomes,in order to provide reference for the effective utilization of flavonoid compounds from Hippophae rhamnoides leaves.The results showed that under the conditions of 75%ethanol,an extraction temperature of 81℃,an extraction time of 110 minutes,and a material-to-liquid ratio of 1∶8 mg·mL^(-1),the theoretical yield of flavonoids was 60.56 mg·g^(-1),with an actual measured yield of(60.52±0.18)mg·g^(-1) after two ethanol extractions.Using D101 macroporous resin as the adsorbent,systematic optimization of parameters such as sample loading,concentration,pH environment,eluent volume,elution rate,and eluent concentration was conducted.The D101 macroporous resin exhibited an adsorption rate of 91.19%±0.32%and a desorption rate of 92.27%±0.75%,resulting in nearly a 2.95-fold increase in flavonoid content,with the adsorption rate surpassing previous reports.
