摘 要:目的研究葛根素对腹主动脉瘤扩张的保护作用并验证其是否通过Keap1-Nrf2信号通路的抗氧化机制来发挥作用。方法选用SPF级雄性健康SD大鼠,随机分成四组,即假手术组(S)、腹主动脉瘤模型组(AAA)、安慰剂组(Pl)和葛根素处理组(Pu)。采用弹性蛋白酶灌注法构建大鼠肾下型腹主动脉瘤模型。所有大鼠均接受手术,假手术组灌注生理盐水,其余三组灌注弹性蛋白酶。术后第二日起连续给药30 d,Pu组腹腔注射葛根素溶液100 mg/(kg·d),Pl组和S组按体重给予腹腔注射葛根素溶液的溶剂作为安慰剂。30 d后所有大鼠再次麻醉开腹,留取动脉标本进行HE、EVG染色,对相关蛋白进行Western blot检测,通过分光光度法进行MDA含量及SOD活性检测。结果葛根素处理有效抑制腹主动脉的扩张,减轻炎症反应,保持弹力纤维连续弯曲的状态,降低TNF-α、NF-κB、MMP-2、MMP-9蛋白的表达水平,上调SM22α、NQO-1、HO-1以及细胞核内的Nrf-2蛋白表达水平,降低动脉组织及血清MDA含量,升高SOD活性。结论葛根素在大鼠腹主动脉瘤扩张过程中发挥保护性作用与激活Keap1-Nrf2通路相关。Objective To study the protective effect of Puerarin on abdominal aortic aneurysm expansion and to verify whether it plays a role through the antioxidant mechanism of Keap1-Nrf2 signaling pathway.Methods SPF male healthy SD rats were selected and randomly divided into four groups,sham operation group(S),abdominal aortic aneurysm model group(AAA,by elastase infusion method),placebo group(Pl)and Puerarin treatment group[Pu,intraperitoneally injected with Puerarin solution 100 mg/(kg·d)].30 days later,all rats were anesthetized again and arterial specimens were collected for Western blot,and MDA content and SOD activity were detected by spectrophotometry.Results Puerarin effectively inhibited the expansion of abdominal aorta,reduced the inflammatory response,maintained the continuous bending state of elastic fibers,downregulated the expression levels of TNF-α,NF-κB,MMP-2 and MMP-9,upregulated the expression levels of SM22α,NQO-1,HO-1 and Nrf-2 in nucleus.The content of MDA decreased but the activity of SOD increased in Puerarin than in model in arterial tissue and serum.Conclusion Puerarin plays a protective role in the process of abdominal aortic aneurysm expansion,which is related to activating Keap1-Nrf2 pathway.
