摘  要:目的探讨常山酮能否通过SIRT1/AMPK信号通路调控关节炎大鼠滑膜细胞凋亡和炎症反应。方法18只雄性SD大鼠随机分为对照组(Control)、模型组(Model)和治疗组(HF),每组6只,对各组大鼠的关节损伤情况进行评分,HE染色观察各组大鼠膝关节软骨组织形态;ELISA法检测各组大鼠关节液中炎症因子IL-6、IL-17、TNF-α的水平;流式细胞术检测各组大鼠软骨组织细胞凋亡率;Western blot检测各组大鼠关节滑膜组织SIRT1、AMPK蛋白的表达。结果与Model组相比,HF组大鼠的关节损伤程度显著降低;膝关节软骨组织形态更为完整;关节液中的炎症因子IL-6、IL-17、TNF-α水平显著降低;滑膜组织细胞凋亡率降低,SIRT1和AMPK蛋白的表达显著升高。结论HF通过激活SIRT1/AMPK信号通路抑制关节炎大鼠模型中的关节炎症反应和滑膜细胞凋亡。Objective To investigate whether halofuginone can regulate the apoptosis and inflammatory response of synovial cells in rats with arthritis through SIRT1/AMPK signaling pathway.Methods Eighteen male SD rats were randomly divided into control group(Control group),model group(Model group)and treatment group(HF group),with 6 rats in each group.Joint injury of rats in each group was scored,and the knee cartilage tissue morphology of rats in each group was observed by HE staining.The levels of IL-6,IL-17 and TNF-αin joint fluid of rats were detected by ELISA.The apoptosis rate of cartilage tissue of rats was detected by flow cytometry.The expressions of SIRT1 and AMPK in synovial tissue of joints of rats were detected by Western blot.Results Compared with model group,the degree of joint injury in HF group was significantly reduced,the cartilage morphology of knee joint was more complete,the levels of inflammatory cytokines IL-6,IL-17 and TNF-αin joint fluid were significantly decreased,the apoptosis rate of synovial tissue was decreased,and the expressions of SIRT1 and AMPK were significantly increased.Conclusion HF inhibits joint inflammation and synovial cell apoptosis in rat models of arthritis by activating SIRT1/AMPK signaling pathway.
